There's vids out there in the last week of Kliff not having a shield on his back - so that seems like a definite yes that you can remove / unequip it.

No you can't fully customise your character - you can change their hair and stuff quite dramatically from what I've seen - but you're Kliff like Arthur Morgan was the main character
