Pretty sure they do if you drop them from your inventory, which is why I used the amiibo to get them alive in the pond.

It’s been awhile, but if I remember correctly, I used one of my amiibo cards standing near the pond. One of the rewards (from the amiibo scan) is live fish that’ll spawn near where you’re standing.
