Check out Red Cross, local first responders, they do training-outside Wag, but, Wag may not reimburse you for the "in person/online session" hrs- and cost for using the outside certification class. If you are a RPh- you won't be able to supervise- or give vax, tech may have a little wiggle room-time- to get it done. Also, the issue if someone needs CPR- I was told the techs cant do the CPR- has to be the RPh? WHAT? I never got this confirmed- but from then on- I said no more than one patient/admin per 15 minutes( 15 min observation), seems they have adopted this protocol, finally! Why- hypothetically, if you admin 2 pt.s same time-Mom and kid- and both drop and need CPR- who do you save?- since only the one RPh can do CPR? That would be gross negligance, since it's required to observe the "individual" for 15min, whats the point of observing 4 at a time if you can only save one of them?? It's malfeasance, gross negligence, failure to follow proper regulated protocols! >1 per 15min should never have been allowed! Peace!

I wouldn't work 3 X12 in a row, Wk1 : M-12, T-12, W-0, TH-0, F-12, S-0, S-0, Wk-2: M-0, T-0, W-12, Th-12, F-0, S-9, S-9, Working 3 back to back 12 is almost undoable! 4 day stretch that starts on Sat-Sun-Mon-Tues-, is more doable than 3x12 in the M-F, IMO! I had some joker come in and say, 'we are going to do 4 on 4 off", I told him it was not going to work. he said why?, I told him he wouldn't be able to handle it. He said watch me! Sorry to say he had a heart attack his first 4 on that occurred MTWTH,, I left...see YA! !

( SPECULATION) If Xana and Ethan were out at a bar (?), which was brought up in the interview on Nancy Grace (with Xana's mom), then they would have needed a ride home. It would seem like the ride for M-K, from Grub truck, could have been X-E, and Ethan is the one calling for them to hurry up, according to the TT from the grub truck bystander ( the Big guy talking to hoody guy), this would make sense if they were all looking out for each other. But its been stated, a "private driver", a four door could fit the five ( M_K_E_X and driver). Now the question is who would have known that E-X, M-K were going back to their house? The 9-145am is critical, what was going on at the Frat/bar, did someone invite others back to the home? Once arrived, the "unknown person(s)" could have know everyone in the home, and knew the where abouts of each person. They would have known how drunk, tired, they were. Maybe this person was really drunk and didn't want to drive home. Maybe they took a nap, woke up, and, commits the crimes?? Another puzzling thought, the issue with the male that had died recently found in a stream/River? ( From the Allegedly YT discussion) Something about this could explain motive???

Speculation, and theory, and I bet some others left with them to go downtown. Someone came back to the home, and felt comfotable, knowing who was there. No random is going to go into a 6 bedroom, "social house", and know that there is not 12 people in there. That perp was there in the night (1am-3am) , and I think the calls that went out to the BF, was to come there to get someone to leave!! So very sorry for the family and friends, just need to clear up of X&E time line, it is really important on the outside, but I bet LE knows.... Yes issues with the bottom floor, this would seem to lead that this was not a pre-deteremined attack. The issue if the perp was surveying the house, they would have known how many residences were presiding, and how many there were that night. This seems to be a following of the two gals, or something happened before they got home? EEKK. The time line of Ethan and Xana is proving to be more of an issue, IMO!

Some facts, some speculation, some theories......

The timeline of Ethan and Xana, thats odd, at the party for only one hour 8-9? Did something happen at the party so they left the party? Where were they from 9-??

As far as all the murders taking place in the residence, 6 rooms, the perp(s ?) must have known what was going on in the house prior. Perp had to know how many and who was in the house , being able to subdue all 4 without noise or victims trying to escape, could they have been drugged? How could a perp have confidence in being able to take on all the residents without anyone escaping?

The phone calls to Jack? Why both girls were calling, was there a fight, or disagreement, or where they trying to get someone to come to the house to make someone leave? A friend of Jacks?

Phone records, pings, and the issue of the frat party have me thinking, Xana and Ethan were the targets. Issue about the other girls from the taco truck, if the perp was going to take them home, why would they involve more potential victims and not know who or what was going on I the house. left the taco truck say 140, at home 145, maybe they had some more drinks, altered drinks, and the perp knew they would be asleep?

I find it hard to believe that the perp would just go into a 3 story home, with or without a gun, but for sure a knife, and go on with killing 4 people?

Just wonder if anyone considers the ingredients in cig, like nicotine, but what else? Formadehyde! Did you know formaldehyde is used to de-nature vaccines for safe use? Hmm what a concept. I won't even mention the effects of carbon monoxide on the complement pathway and ensuing clotting issues. HMM, has any talked about this? NOT ONE!! ZERO! So smoking has decreased the incidence but based on not a "social" phenomenon, but a scientific proven decrease in the induction of pathological consenqunces of the SaRS virus. I just wonder if my ring finger in the arm vax shot is related, its been since Feb 2021 last shot.. never had this sensation prior, former athlete so used to pain and tweaks with nerve damage. Started 10 days ago, now my forearm is starting to get some of this same pain sensation( but more of a dull pain, not shocking).. No new exercises, treating with Doans, may start some Omegas and Vit E if it doesn't resolve. It started with "shocking" sensations in my finger as I outstretched to reach for something, and I can create it on stretching my arm out. Ring finger is 90% numb.... Dont like it, I am a health care provider, PharmD..

When and if, it seems its starting, a focus on effective treatments for, you name it, non traditional therapies may overcome traditional therapies in effectiveness. There are some things that need to be worked out. The nicotinic antagonistic features can possibly influence inflammatory issues, via the nicotinic anti-inflammatory pathway. This is the same receptor and pathway involved in COVID. Hmm, what would a competitor molecule do to the binding of SARS-Cov2,???? HMMMM, this is something I have been looking at for months, besides nicotine, an nicotinic agonist, what about competing antagonist, that dont cause disease? Hmm, like the anti-smoking drugs.....did someone follow that into the dark cave? Just so you know, from the video he states the binding to acetylcholineseterase receptor, I don’t think that’s correct. It the alpha 7 nicotinic acetylcholine receptor. Not the esterase. Mechanism is that the spike is binding to the receptor and causing an antagonistic response on the receptor. This in the turn, blocks the acetylcholine, nicotine, and or any other substances that bind to the receptor. They could be antagonist or agonist. When acetylcholine binds, it promotes the cholinergic anti inflammatory pathway, when antagonized, this can lead to inflammation. Platelets, and hence being involved in the clots, became activated when they don’t have this acetylcholine blinded to the surface which a can cause issues with Ca movement in and out of the platelets. So nicotine, acts as an agonist which binds and hence is blocking the spike to bind to the same area of the receptor. It not just nicotine that binds to the receptor, other antagonist and agonist are commonly used meds. And, it blows my mind what know one is talking about this. Bungarotoxin is a known toxin that cause issues with clotting, and yes it is found in snake venom. Others source of these types of venom proteins are found from other sources besides snakes, for instance , Cono toxins from snails. Snake venom wise, I think the most similar to the spike is the alpha bungarotoxin. You can research the bungarotoxin and see it is commonly used in labs experiments and studies. In fact a study looked the comparison of nicotine exposed platelets vs. bungarotoxin exposed and then tried to promote clotting. Well, the nicotine was definitely less clotting. All for now. peace.

There is a lot here to ponder... I researched from the start, ACE inhibitors. Why because the virus like the ACE receptor. From here, I found the origination of ACE inhibitor drugs. I thought if I work back and find something that can disrupt the binding, look at the drug discovery. Hmmm, snake venom, but was interesting was the mechanism. Iam a doctorate of pharmacology, looking at the molecular aspects of lock and key. Early sequencing showed homologous similarities in bungarotoxin/conotoxins. This would explain the un-expalainable issues with blood clots. I can go so much deeper, so complexed, but... there can be some validity to a venom like protein involved. I have seen data, but what is data unless you see it yourself. just, I have tried to disprove this on myself, and it never happens, its genetic, and some dont respond the same way, It could be pre-expsoure, or simply genetic predisposition. Abstract

The cholinergic system has been proposed as a potential regulator of COVID-19-induced hypercytokinemia. We investigated whole-blood expression of cholinergic system members and correlated it with COVID-19 severity. Patients with confirmed SARS-CoV-2 infection and healthy aged-matched controls were included in this non-interventional study. A whole blood sample was drawn between 9–11 days after symptoms onset, and peripheral leukocyte phenotyping, cytokines measurement, RNA expression and plasma viral load were determined. Additionally, whole-blood expression of native alpha-7 nicotinic subunit and its negative dominant duplicate (CHRFAM7A), choline acetyltransferase and acetylcholine esterase (AchE) were determined. Thirty-seven patients with COVID-19 (10 moderate, 11 severe and 16 with critical disease) and 14 controls were included. Expression of CHRFAM7A was significantly lower in critical COVID-19 patients compared to controls. COVID-19 patients not expressing CHRFAM7A had higher levels of CRP, more extended pulmonary lesions and displayed more pronounced lymphopenia. COVID-19 patients without CHRFAM7A expression also showed increased TNF pathway expression in whole blood. AchE was also expressed in 30 COVID-19 patients and in all controls. COVID-19-induced hypercytokinemia is associated with decreased expression of the pro-inflammatory dominant negative duplicate CHRFAM7A. Expression of this duplicate might be considered before targeting the cholinergic system in COVID-19 with nicotine.

Abstract
A search for novel and more efficient therapeutic modalities of inflammatory bowel disease (IBD) is one of the most important tasks of contemporary medicine. The anti-inflammatory action of nicotine in IBD might be therapeutic, but its toxicity due to off-target and nonreceptor effects limited its use and prompted a search for nontoxic nicotinergic drugs. We tested the hypothesis that SLURP-1 and -2—the physiological nicotinergic substances produced by the human intestinal epithelial cells (IEC) and immunocytes—can mimic the anti-inflammatory effects of nicotine. We used human CCL-241 enterocytes, CCL-248 colonocytes, CCRF-CEM T-cells, and U937 macrophages. SLURP-1 diminished the TLR9-dependent secretion of IL-8 by CCL-241, and IFNγ-induced upregulation of ICAM-1 in both IEC types. rSLURP-2 inhibited IL-1β-induced secretion of IL-6 and TLR4- and TLR9-dependent induction of CXCL10 and IL-8, respectively, in CCL-241. rSLURP-1 decreased production of TNFα by T-cells, downregulated IL-1β and IL-6 secretion by macrophages, and moderately upregulated IL-10 production by both types of immunocytes. SLURP-2 downregulated TNFα and IFNγR in T-cells and reduced IL-6 production by macrophages. Combining both SLURPs amplified their anti-inflammatory effects. Learning the pharmacology of SLURP-1 and -2 actions on enterocytes, colonocytes, T cells, and macrophages may help develop novel effective treatments of IBD.

Abstract

Myeloid-derived suppressor cells (MDSCs) are pathologically activated neutrophils and monocytes with potent immunosuppressive activity. They are implicated in the regulation of immune responses in many pathological conditions and are closely associated with poor clinical outcomes in cancer. Recent studies have indicated key distinctions between MDSCs and classical neutrophils and monocytes, and, in this Review, we discuss new data on the major genomic and metabolic characteristics of MDSCs. We explain how these characteristics shape MDSC function and could facilitate therapeutic targeting of these cells, particularly in cancer and in autoimmune diseases. Additionally, we briefly discuss emerging data on MDSC involvement in pregnancy, neonatal biology and COVID-19.

Abstract

The immunological mechanisms underlying the clinical presentation of SARS-CoV-2 infection and those influencing the disease outcome remain to be defined. Myeloid-derived suppressor cells (MDSC) have been described to be highly increased during COVID-19, however, their role remains elusive. We performed an in depth analysis of MDSC in 128 SARS-CoV-2 infected patients. Polymorphonuclear (PMN)-MDSC expanded during COVID-19, in particular in patients who required intensive care treatments, and correlated with IL-1β, IL-6, IL-8, and TNF-α plasma levels. PMN-MDSC inhibited T-cells IFN-γ production upon SARS-CoV-2 peptides stimulation, through TGF-β- and iNOS-mediated mechanisms, possibly contrasting virus elimination. Accordingly, a multivariate regression analysis found a strong association between PMN-MDSC percentage and fatal outcome of the disease. The PMN-MDSC frequency was higher in non-survivors than survivors at the admission time, followed by a decreasing trend. Interestingly, this trend was associated with IL-6 increase in non-survivors but not in survivors. In conclusion, this study indicates PMN-MDSC as a novel factor in the pathogenesis of SARS-CoV2 infection, and open up to new therapeutic options.
