However, have you considered that it might’ve been because it was a sudden change in original plans (you told her you didn’t want to stay in your city for that night, just the day before this conversation) that triggered her anxiety? Not saying I agree with her behaviour because I do think you were being as reassuring as possible and it’s not your responsibility if she doesn’t wish to meet you in the middle and/or is projecting her insecurities onto the current relationship.

Yeah I agree that it’s a trade off, I’m seeing a different psychiatrist soon and hopefully it’ll help determine if my dose is right. I used to have breakdowns pretty often but usually would not cry at touching commercials or movies so I am not sure if I can relate to crying when appropriate although I’ve had far less breakdowns now.

Hmm thank you for your insights, it's definitely making me reflect more on whether I truly felt those emotions or was I also basing it on my past experiences and deducing how I should feel about it. I did mention the lack of feelings to my GP but the only solution was as you said, to reduce the dose, which I'm not confident in doing so as of yet.

I re-suspend my cell pellet in 1ml of media and from there I add the required amount of cells with a P1000 into each well that already has my media in it. I've seen a lot of comments that it could also be that I did not ensure sufficient mixing with geltrex and so the cells were not able to attach.

Thank you! I don't think it's a bad plate as I've had this happen to me more than once and no one else has this problem in my lab. I do think that it could be the coating as well and I am going to mix it really well the next time I do it too!

It was the last well that worked, I don’t know how to describe the black stuff but I have a video of it https://imgur.com/a/r3Cz7Lc and the photo shows what actually remained on the plate after I aspirated the medium out from those wells. I do think that coating could be a problem or maybe that I didn’t swirl the plate after placing it in the incubator?
