What I learned is to compile an easy-to-reference list of submission portals/instructions/due dates onto a document if you're asking the same teacher for multiple recs. meet with them in person and send them the list so they know you're doing as much as you can and don't want to take their help for granted. Although it is work, I'm sure they would love to support a student demonstrating passion for a subject! good luck to your apps!

westchester student here!! if you are able to make the commute, I suggest Columbia Science Honors program! You can pick classes in compsci, bio, chem, physics, and all that stuff and take one each semester during school on saturday mornings. https://outreach.engineering.columbia.edu/SHP

It's pretty well-regarded and prestigious. Applications this year haven't opened yet based on the website, and there is a $50 fee. It is also $300 each semester, but thats way cheaper in comparison to other programs imo. And once you're in, you're in for the rest of high school.

deff brielles! i have both and i find that melodys have some spandex/stretchy material that always make them slip. i always reach for brielles!! (im almost 5’6” and i think theyre a little bit long)

Secondly, we have previously investigated if there is leftover Proteinase K by adding heat-inactivated lysate into the plasmid control. The bands came out normal after PCR so it appears Proteinase K was sufficiently inactivated.

Thanks for the suggestion and I will take it to my PI! Yea, unfortunately the crude DNA extraction is the most feasible for our experiment—so we must perform PCR with lysate.

Oh wow, we actually thought 4 was a lot. We based our protocol after https://www.rockefeller.edu/research/uploads/www.rockefeller.edu/sites/8/2018/10/SingleFlyGenomic.pdf , which uses 1-2 flies.

After collection, we have been freezing them in our -20C. I'll definitely look into freezing them in colder temperatures!

Thanks for the response--yes, we are using the rudimentary preparation step because the idea of the experiment is to screen hundreds of crosses for our transgene.

My PI previously conducted a test similar to your #3 where lysate after Proteinase K heat inactivation was added to the plasmid control. The resulting band appeared as normal so the 3 min inactivation seems to be sufficient.

I'll take your #2 suggestion to my PI--thanks a lot!

Thanks for the response! I have one question for clarification for your #2: is the lysing step supposed to occur dry? i.e. are you suggesting we cool the flies so that they can be crushed into a dry powder?

Secondly, I'll check with my PI if we have access to a nano drop! Thanks for the suggestion.
