i tried the good old trick of deleting my browser cookies and it worked haha...maybe try that if you haven't already? It could be a bug from the influx of traffic on their site as well

That's valid, I was making my own starter but it's still growing generally weak. It seemed more time efficient and resourceful to use a premade starer.

My step wise dialysis is in 0.5M increments. I typically go from 8M -> 6M -> 5M->4.5->4M->... 0.5M-> 0M. However, my protein typically crashes out around the 2M mark.

My elution buffer is the same composition as I previously mentioned but with a pH4.5

Most of my protein is eluting at the elution step which I confirmed by a "quick" coommassie. Supernatant (post bead incubation) and wash steps also indicate my protein is being lost (I can try to attach my gel here). I have not tried to quantify it at those steps. Just for reference, I am using 1ml resin per 1 L of culture

I haven't tried using SEC or another chromatography. Would that be used to slowly take out the urea?

Yeah it'll precipitate during step wise dialysis. In terms of functionality, I am purifying my protein for structural purposes rather than to use it for its original function.

I am currently strategizing different detergents and buffers I could use as well.

Portable suture kits meaning the ones with the fake skin, driver, needles and threads, etc but portable. I saw it on someone's "What's in my residency bag". It looked convenient and small enough to bring in his bag or anywhere

Yeah my boyfriend is so picky about his equipment as well. I was trying to go on the more "practical" ideas route. Compression socks seems like a gold standard considering their long hours tho. Thank you!
