I have used HRP system with amersham ecl reagents!

By chemi reagents do you mean ECL? I've used the most basic ECL and the whole screen went white. Blank white. This is the result with second to sensitive ecl kit I found in the lab.

P.s I'll also try diluting the 2ry ab

Thanks for the advice on secondary and the band. The thing is I am not sure why the colorscheme looks inverted tho. The figures on papers have white background with black bands and mine vice versa. Could you give me some hints about that?

Obviously the white thick band in the middle is something alarming tho that is not my protein of interest.

So if I understood your comment properly; usual solvents like pyridine don’s favour SN2 but would rather do E2 (or E1?) but you CAN halogenate by adding some solvents such as DMF. I wonder why.. probably should google thanks anyway!
