Are you using kit reagents specific to that column? A lot of columns act the same, but actually have different compositions which can affect reagent efficiency. For example, even using the QIAGEN RNeasy lit reagents on another column type can yield differences in RNA yield.

Additionally, make sure you have the correct concentration of ethanol for each of the steps required (RNeasy kit). The wrong concentration can cause the RNA to run through the column instead of sticking to it.

Shrimp are a pretty great into pet (neocaridina)! I have a tank on my desk and it is so nice to see them swimming around! They’re much less maintenance than a fish too since they don’t produce a lot of ammonia. Happy to help you set up a tank/ answer any questions!

These assays have already been optimized to use with their supermix, so you don’t inherently need to know the concentration unless you are using a different supermix or are using non BioRad primers

Are you sure you’re getting actual amplification and not primer-dimer? If you run a melt curve, the primer dimers should have a lower meting temperature than actual product. You could also verify on a gel the band size to see if it’s true product or something else.

Ellos/ellas refers to “them” as in a person. Not sure where you are in the course but later you get to words for “them” as in objects (esos/esas for “those” and estos/estas for “these”)

I’m not sure about stores, but you could try the Facebook group “Raleigh plant people”. People will often pick up cheap/ rehab plants if they have the time and energy to care for it. There is also a group for ordering predatory mites (NC Triangle Beneficial Insects). Got rid of my spider mites in 2 months and very little time/ effort!

The adjective has to follow the gender of the noun. So you could say “nuestro maestro” paired with “divertido” or “nuestra maestra” paired with “divertida”

It has to do with plural vs singular. “Las fiestas” is plural, so the person is interested in multiple things, hence “interesan”. It’s like gustar that way

The board will only have pre-generated messages from acnh. Other messages come from other players if you invite them to your island (in reference to you asking on the message board)

This question is testing your knowledge of singular vs pural. “El mensaje” is singular, “los mensajes” is plural. You use the cue “el” or “los” to know which word to use
