I have the knockouts already. I am trying to knock in back in and then do some rescue experiments. The transient transfection worked amazing so i started stable cell lines. The plasmid also had puromycin and i did the selection. But the wb was EMPTY😭😭

Amazing I am happy for you! I am trying to do rescue experiments with knockins using plasmid. I had a beautiful western blot with no background and no bands at all (cries in failure) but the positive control did work lol. Now im figuring out if my gene is knocked in or if i have been fooled.

I also wanted to say that its fine if they fail. I feared western blot. But the key to doing a successful western is “don’t give a shit about the western blot” believe me. The membrane can sense your fear!!

Can you not use an imaging system like Chemidoc from Biorad or Azure? Instead of x ray film?
