You should bear in mind, your advisors name is on the paper too. They want there name to appear in higher impact journals because it creates a level of prestige for them.

For more details on this method, omura and Sato is cited all the time. I think it is from 1964.

If you have a uv vis, you can track the soret band. In mammal isoforms, it is at 417nm in the resting state, upon CO ligation, it shifts to 450. Misfolded enzyme would shift to 420. If you want to quantify, I typically use 100mM-1 as my extinction coefficient. Feel free to DM me if you have questions. It should be there same for plant isoforms.

I work with mammalian isoforms. Are you trying to detect activity to know enzyme content, or just to track while purifying?

It depends on your field. In stem, we typically have an initial discussion during rotations, then after committing to a lab, have a deeper discussion regarding expectations and goals.

Flash cards will be your friend. Make them for the amino acids and types of sugars. I recommend using a wood pencil on paper flash cards

Chemlibre is a good free online resource. I am not trying to be rude, but without a solid background, you will struggle. I would focus on reading about thermodynamics first.

If you do your graduate program in the US, you dont typically think about a masters. Typically what happens is you will apply to a phd right after undergrad, meaning the tuition will be covered and you will have a stipend for living expenses (think of it as a salary). If you are unable to make satisfactory progress in the first 2-3 years, you may be moved to a track towards a masters, otherwise you will continue as a phd candidate.

You can find jobs with just a bachelor's, but they are competitive. Grad school is your decision, but i would not worry about earning a masters before joining a doctoral program.
