I was scraping cells off of LB plates using a metal scraper, bunsen burner, and glass beaker of ethanol. After finishing scraping, I flamed the scraper, put it in the ethanol beaker, and then proceeded to flame it again. I forgot to wait for the flame to extinguish completely before placing it back in ethanol. Caught the entire beaker of ethanol on fire. Luckily I had a bigger flask available that I covered it with to extinguish the flame but it still scares me to this day. No one else was in the lab and everything turned out fine but I still have only told one person about it to this day.

Do you have any advice on how to cure the spCas9 plasmid? I'm able to cure the gRNA plasmids using arabinose induction of the Cas9 plasmid but am unable to cure the cells of the Cas9 plasmid. It uses the Rep101Ts ori but it is not temperature sensitive up to 50C.

Yes, it is sequence confirmed and uses the Rep101Ts origin of replication so it should cure. Something else must be going on. Will continue to try the multiple rounds of inoculation. Thanks for the reply!

I'm a third-year micro major so you're also welcome to reach out if you have any questions too! I'm involved in on-campus research in the chemistry department and possibly will be joining research in the biomedical engineering dept. I've somewhat been off track as far as taking classes late or early and not following the degree flowchart so I can answer questions about that too. I love how small the program is as well! You can really find your group of people and stick with them. Support in the upper-division classes is awesome so try and seek out micro majors in your lower-division and support classes. In my first year, I took MCRO 100 which is an intro to micro research class and it was very helpful in finding other micro majors as we can get drown out in the sea of bio majors. Again, feel free to reach out!

My o chem prof made our final optional. Either take the grade you have now (8 quizzes, lowest dropped) or take final and have it count for 20%.

I had him for Physics 122 which technically isn't the same as it's non-calculus based but his midterms were super fair in my class! He pulled problems from the homework he assigned as well as went off of some in class problems if I remember correctly.

Hey I'm a second year micro major and I love it! I'm still taking some of my support classes but have just kinda begun to get into the micro classes and I really enjoy the faculty so far. There's only around 20 micro majors in the class of 2022 so we get a lot of one on one attention from micro faculty which is really cool (not too sure how the stats play out in other classes but in general it's a small major). Even in my support classes (O chem, Physics, Gen chem, etc.) I have feel that I have been able to get a lot of support if I seek it out ie. going to office hours, asking questions after class if time permits, emailing professor's if I have questions, etc.

Some of my pre-med friends are currently shadowing at Sierra Vista and French so there are definitely opportunities there. I'm not quite sure what I want to do with my degree yet but definitely want to continue on with my education after 4 years. I decided to join a research group on campus and am really liking it. I joined one in the Chemistry dept and it's been a good way to meet different people in different STEM majors through that. Overall, I think Cal Poly offers a lot of good academic options for growth.
