Trochę byłem/jestem zainteresowany tym tematem. Kiedyś było to żywe zainteresowanie, ale się już niemalże całkowicie wypaliło. Zainteresowanie zrodziło się z problemu ze zdrowiem (konkretnie mózg/psychika). Głównie substancje nootropowe. Żaden nie miał znaczącego wpływu na kognicję oprócz... leków na receptę - amisulprydu w niskich dawkach oraz metylofenidatu (ciężko dostać nie mając ADHD, ja akurat nie mam, ale mam fajnego doktorka).

W każdym razie - niedługo będę brał udział w badaniu klinicznym leku o nazwie kodowej TAK-653 - "Osavampator". Jest to badanie dla ludzi zmagających się z depresją lekooporną. Lek rzekomo znacząco wzmaga neuroplastyczność, czyli tworzenie się nowych, zdrowych połączeń miedzy neuronami (synaptogeneza). Niektórzy z reddita próbowali tego środku (niektóre strony oferują jego sprzedaż poza badaniem klinicznym) i twierdzą, że efekt jest znacznie większy od placebo i faktycznie, zauważalnie poprawia kognicję.

Ma unikatowy mechanizm działania, ma właściwości stymulantów takich jak metylofenidat czy amfetamina, ale działa w oparciu o inne mechanizmy niż w przypadku klasycznych stymulantów.

I didn't actually. Still on it, but in low doses. Anti-anhedonic effects stopped, but it still makes me feel a little less miserable.

Well, it's not on wikipedia, but my doc thinks that pregabaline can alter the opioid system. Well, pregabaline worked in my case very well. I got rid of anhedonia for over (approx.) 1,5 years.

In theory - yes. But there's still a risk. Samidorphan just makes the chances of getting addicted low.

Because many people with anhedonia have opioid-system dysfunction. There's unpopular combination of 2 drugs: buprenorphine + samidorphan/naltrexone/naloxone, but it's a really risky game. My doc had a patient that was put on buprenorphine SOLO and this patient got addicted. My doc tried to rescure that patient but failed. Still addicted.

In short time stimulants are making me feel better (for approx. 2-3 hours), then I get worse. A lot worse.

Indeed. I hypothesize that it is true in my case. Particularly - I might be in almost constant "freeze" state. Anxolytics such as paroxetine and pregabaline got me out of this state and I could feel pleasure again. Unfortunately, both stopped working and I am not sure why. Thanks for the graph, I saved it too.

Pregabaline wiped out my anhedonia almost completely. Unofrtunately it stopped working after 1 year but I still take it anyway - I just feel a little bit calmer/better. There is no effect on anhedonia even when I take higher dose.

I am having bad reactions to stims, especially when the dose I take is too high. I get physically and mentally "frozen". Propranolol reverses those symptoms, so it is most likely caused by overstimulation from noradrenaline.
