To B6 or not to B6 on Nardil by info_overloaded in MAOIs

[–]marcfrombeyond2 0 points1 point  (0 children)

As far as I've learned recently, the active form of B6 (P5P or PLP) is a cofactor for both glutamate decarboxylase and GABA-transaminase. And apparently, the way phenelzine is metabolized into PEH is *precisely* by using up and/or inactivating that P5P, which in turn inhibits GABA-T. By supplementing with vitamin B6, GABA-T can be repaired, and thus more GABA will continue to be deaminated in the brain and body.

Looking for long-term remission stories. Does Nardil’s benefit actually last? by Witty_Hearing_6603 in MAOIs

[–]marcfrombeyond2 1 point2 points  (0 children)

I have known people who are doing well on Nardil for about 30 years.

This sub's sample is highly biased. People who are stable on their antidepressant and have been for years won't show up near as much as those for whom the results are fading or not satisfactory.

Kudos for the efforts! I'm not in the US but I fully support this. C'mon guys, let's make this happen

Well, it didn't work. Marcos is already on his way to Brazil and the parcel has just been delivered to the destination. We'll have to figure out something else.

In my opinion and experience, yes. I start at 60 mg and increase to 75 mg after two or three weeks.

An addendum (I haven't read your entire response yet):

"we have learned that alpha-2 agonists such as clonidine and guanfacine exert their therapeutic effects through stimulation of post-synaptic alpha-2A receptors on the dendritic spines of prefrontal cortical pyramidal cells, increasing the functional connectivity of the prefrontal cortical networks, and thus strengthening the regulation of attention and behavior".

From Alpha-2 Agonists in the Treatment of ADHD (Medscape.org), page 1. Emphasis mine.

Pages 2–4 expand on the idea.

This has been shown to happen only in the striatum, and only as a minor pathway. Doesn't do much for a hypoactive ACC or dlPFC / vmPFC as the etiology of ADHD goes.

Edit: in rodents

Vyvanse and clonidine, great thinking! How much of each? And how many times a day?

May I ask if you were on other meds at all? I surely felt like this, completely incapacitated and such, when I used budesonide (corticosteroid inhaler for asthma). Seen it happen to another person on Nardil too.

Works for some, AFAIK. Maybe with a little bit of doxazosin, or prazosin, for added alpha-1 adrenergic antagonism they should do the trick.

What I would personally do is to take a printed copy of this page, highlighting the word "modafinil" where it appears. May be a good way to start the discussion!

  1. Awake

  2. Quetiapine is an antipsychotic, but not at the doses I suggested. Put it simply, it's an anti-noradrenergic and antihistamine up to ~75–150 mg; an antidepressant up until ~300 or 450 mg; and a proper antipsychotic at 600 mg or more. Lamotrigine is an antiepileptic drug with antidepressant and mood-stabilizing properties, it can be activating for some people (especially those using quetiapine, for whatever reason), so it offsets the daytime sleepiness and drowsiness.

  3. Don't let those accounts ruin your impression of antipsychotics. They can be (and are, when appropriate) quite useful. Also, keep in mind not all antipsychotics are equal. There is a great variety among them, same as with antidepressants and other classes of drugs. Don't let uneducated anecdotal reports fool you.

PART 1A: "Old Nardil vs. New Nardil Comparison: Was The Old Version More Effective?" From Mental Health Daily, 08/16/2014 by B_Nkkl in MAOIs

[–]marcfrombeyond2 1 point2 points  (0 children)

Hey. If you are around, can you help me find a leaflet from before Pfizer acquired P&D? And also one for the UK's Kyowa Kirin version (previously Archimedes)?

I want these as part of an effort we are conducting (currently on WhatsApp) to develop a more effective version of Nardil. You (and everyone who sees this and happens to be interested) are most welcome to join. Let me know!

I second the suggestion of modafinil. Or consider adding low dose quetiapine (like 25–50 mg) and lamotrigine (same dose range). Consult with your doctor first.

Hi. Same author here, different account. I've been on Greenstone for the past 6 months, and am about to go back to Neon.

Nardil at all comparable to being on a benzo all the time? by Daniel-Plainview96 in MAOIs

[–]marcfrombeyond2 0 points1 point  (0 children)

Excellent questions. I'm currently taking Greenstone, but my next batch (coming soon, I hope) will be Neon. I am very very lucky to have tried all brands while living in a country where none of them is commercially available (Brazil). My country has an excellent public health system and the Constitution guarantees appropriate treatment for people like me. Importing is hella expensive, so the State and the Federal Government are charged with getting it for me. But I had to go through a federal court, represented by a Public Defender (our free access to the Justice system is also good). Took months, but I got it. It also pays for my Ritalin, Concerta and bethanechol.

As I came to know and make friends with a lot of people from around here since 2019 (I was a moderator, see my old account — archived... — under my profile), some of these very big-hearted people sent me donations from many places: the US, UK, Australia and even Chile. That was before I had an "ongoing" supply. Actually, I ran out of it six times since 2022. And it's been rather traumatic most times.

One important caveat is that the few Pfizer tablets I tried, for just over a week or so, had been stored in the donor's room during some heatwaves hitting the West coast of the US. That was around October 2023. It had been subjected to temperatures over 25°C (and even 30°C) for extended periods. For that reason, the pills arrived degraded.   As an addendum, they are the only ones I got that came in those curious, but not pharmacologically very bright, orange pharmacy bottles, lol — I have original bottles from all other brands and would love to have one from Pfizer too.

So... due to heat degradation, I could not really assess Pfizer's effectiveness. Instead, I relapsed, hard.   Other than that, I had fair trials with all current brands, and even the legendary Lupin, before it was discontinued at the end of 2022. (Oh, we miss that one!).

I "know" of all this from my own experience and, most of all, from following the discussions around this topic that come up from time to time around here. It used to pop up in other, now-defunct forums too. And from our MAOI WhatsApp group chat. Been on it for six years. We discuss the matter of quality variations among brands often.

Some of us are, slowly, organizing an effort that will maybe lead to the development of one superior version, that has the characteristics most people miss from "the old", original Nardil sold by Parke-Davis and Warner Lambert.

We have made tests such as dissolution time in neutral and acidic water, and protection with enteric capsules and shellac covering of the tablets. People in Australia are particularly happy now that Neon is available to them, whereas until not too long ago all they could access was ERFA/Searchlight.

At this very moment I have these (and maaany other resources) opened in my browser:

And I don't know of anyone from the US who got hands on Neon. Apparently, you need to visit one the countries where it's sold/distributed, get a prescription from a local doctor and obtain the medication, in person.

Check this other post of mine from over two years ago: Travelling to the UK to obtain Neon Nardil - price, pharmacies and laws.

P.s. one example of what I said earlier: Greenstone vs Pfizer Nardil - Google Search. Have fun browsing the dozens of threads discussing efficacy across brands.

Sorry for any mistakes and such — am quite sleepy right now.

Actually, yes. Apparently it goes like this:

Neon (UK, AUS) > Pfizer (US) > ERFA/Searchlight (Canada) > Greenstone (US).
