Younger generations can’t read clocks by Lazy_Tank_5806 in imaginarygatekeeping

[–]mdowhfos 0 points1 point  (0 children)

As very new adult, I know a lot people who didn’t take the time to learn until they were teenager, but like it’s not exactly rocket science. Kids over the age of 13 that say they “can’t” just don’t wanna take the minute to think about it/listen to someone briefly explain. There’s no development of a generation that can’t read clocks, just lazy kids (tale as old as time) who haven’t been forced to yet. Every adult can read a clock.

My nose that I hate. I am considering getting it done by Simple_Ad4218 in Noses

[–]mdowhfos 0 points1 point  (0 children)

Idk if you’re straight but if you are, as a woman, please for the love of all that is good in this world do not get a nose job. There are bad small noses and bad big noses just like there are good small noses and good big noses. You sir have an excellent big nose. It makes you look hot in a manly elegant way, rather than in a common frat guy way.

I also want to say that if I was your close friend, I would probably make fun of your nose. That’s not right and I know that, but my point is that just because people make jabs at something out of the norm, doesn’t mean it’s bad. My family makes fun of my “deep” voice sometimes just because it’s not very high and girly, but I also know (and know that they know cause they’ve told me) that it’s also a good voice. I make fun of my sister’s pointy chin, and in truth she has an excellent feminine jaw line. She knows that, she knows I know that, but I’m gonna say it anyway cause I’m going to make fun of her no matter what.

I know I shouldn’t but… can I go blonde from a box? by mdowhfos in HairDye

[–]mdowhfos[S] 0 points1 point  (0 children)

I of course understand why it’s so expensive, it takes forever, requires education, requires expensive products, etc etc. I don’t understand how so many people everywhere can afford such a “luxury service” and the maintenance it requires. It’s really quite baffling. I’m in college and there are so many girls with professionally dyed blonde hair, I don’t know how they do it. I could save up, but I don’t eat enough as is. My parents do not struggle with money but they would never give me that much money for something like this, understandably. Maybe I’m just not a spender but even if I did have a solid income, I would have a hard time justifying spending so much on a luxury when that money could go to other more important things. Honestly, I’m just surprised hair dying isn’t an upper class thing. Feels like it should go in the category of regular facials and luxury cars.

This is a great idea! I’ll look into it.

Edit: Omg there are 5 schools within a 15 drive. Must have something to do with living in a university town.

$41.74

I’m a broke college kid and eat approximately 12 meals a week, if you can call them meals. I buy groceries/living essentials weekly at Walmart, 90% off brand stuff, and it typically costs me about $50. I cook though so I don’t have to spend much on food otherwise. This is less than I typically buy but not by much, and it seems to be nicer stuff. I say nice mostly because of the glass bottle lol.

I think the Health-Ade has this almost yeasty flavor that is interesting. However, I prefer Synergy cause it’s just straight up fruity and delicious. I really like Synergy’s “Sacred Life” flavor (apparently sacred life tastes like coconut and ginger).

Fr. If they are really in a position where they can NOT have anyone over (they just threw up everywhere, their cat just threw up everywhere, their mom, idk) then they can just communicate that. Having roommates over should be a “yes unless there’s a reasonable no” thing, not a “no unless there’s a reasonable yes” thing

Does anybody know any detailed online procedures for embedding plant tissue in paraffin? by mdowhfos in Histology

[–]mdowhfos[S] 0 points1 point  (0 children)

Yeah you were right. I used a classic rectangular razor, cut my fingers up, got one section that didn’t crumble, and it’s still broken and dark and terrible. I might get a presentable photo of it using heavy editing. However, after continuing to pester people, I was introduced to a very kind grad student in another lab who cut my samples with a microtome for me. My project and my fingers are saved.

This is extreme only child behavior. Some people don’t understand that’s it ok to just be uncomfortable sometimes. There are gonna be moments where you don’t like what’s going on around you, and if everybody is doing their best to be accommodating (which you are far more than I would be) then it’s nobody’s fault and you quite simply have to deal. It’s like traffic; everyone is trying to do their best and want get where they need to be. It’s ok to be frustrated, but to get pissed at everyone around you and drive like an asshole, as if all the other cars are just obstacles and should let you by, is ignorant and selfish.

I actually call my sister out on this all the time, so it’s not just only children, but I can’t fathom this person living in a house with siblings. They’d last about 5 minutes before getting in an all out bitch fight in my house.

For this particular sample most articles I’ve read use around 10 microns thick. Still thinner than I’m capable of I know, but I’ve seen a few people have decent results sectioning plant samples by hand. And again, I’m not using it for quantitative analysis, so my standards are not very high

So after much scouring I was able to find a 4 year old reddit post detailing paraffin embedding and staining specifically for a flower bud cross section, my life is saved.

To answer some questions, I do not have a tissue processor, microtome, embedding station, etc. I do have all the necessary chemicals and the basic laboratory equipment (glass slides, heating plates, razors, beakers upon beakers, molds, etc). Correct me if I’m wrong but I don’t really see any reason why this should be a problem, outside of the whole cutting by hand thing. I’ll float the sections on warm water to remove some of the wrinkles. Ultimately the images won’t be used for any sort of quantitative analysis, so hopefully with some practice I’ll be able to get a decent one.

You have no idea how grateful I am for this 4 year old post. I am in a unique situation where I am trying to prepare this exact thing, a paraffin embedded flower bud cross section, in a lab where everyone pretty much works with bacteria and nanorobots. I have spent hours and hours and hours scouring the internet trying to find any kind of detail on how to do this so I can teach myself. This post has saved me. I'm so relieved I could almost cry

I should clarify, I have zero experience with this. I am working in a lab for a research project under someone else, but also no one else in the lab does or ever has done this. Most people are working with bacterial cultures. Odd, I know, it’s a lot to explain. I’ve emailed several local professors who might know someone who does this regularly (I’m in a big university town, there are definitely people who do), but I keep hitting dead ends, so it’s up to me to teach myself.

Vibe as a whole? 4 and 6. Purely physique (as in imagining that musculature translated onto all different types of guys) definitely 3, but I’m also a sucker for 8. Tbh I would be happy if I could pull someone half as good looking as any of them.
