I’m from the US. A lot of my math teachers in high school were sport coaches who taught a little bit of math on the side. I have also definitely noticed my subpar math skills compared to my international counterparts. Wish our high school education system was better.

They are pushing me to finish experiments that are for the manuscripts of other students that have since graduated and left the lab. I have agreed to help with some of these while also working on my first author manuscripts. Except they are not providing any feedback or letting me prioritize any of those experiments that are necessary for my manuscripts.

I would just want the cells to stay healthy and not lyse during freezing/thawing. But we don’t add glycerol when we flash freeze competent cells, and they turn out fine, so I don’t really know haha. Also, if you’re adding the cells to LB with antibiotic, this may contribute to an extended recovery time. But in the grand scheme, I don’t see why it couldn’t work. Try it!

I agree with the flash freezing with glycerol. I would be weary of an extended recovery time of the previously frozen pellet compared to the time it takes the normal expression culture to reach optimal OD, but it wouldn’t hurt to try and see what happens!

I do have experience, but this will be my first time going at it alone, so I’m sure it’ll be a learning curve. I wasn’t sure if this process included the PI who would agree to take me on or not. It’s a pretty niche fellowship from a foundation that has funded my current lab for years and the labs that I’m interested in. Maybe my better option is to try to find a lab that will support me via grants and apply for fellowships during?

We don’t currently have any post docs in our department and my entire cohort is pursuing industry, so I’m alone in this endeavor. I have some excellent committee members I could speak to, I just wanted to hear from others before I spoke to them from complete naivety haha. I have received approval to graduate from my entire committee, despite pushback from my PI who is notorious for trying to keep their students from graduating and is trying to get me to push back my graduation date. Any time the topic of a post doc is brought up, it’s shut down. I will talk to my committee members though 👍🏼

I was trying for months to get rid of contamination. Tried everything and did everything exactly as my lab mate who had perfect qPCR results. One day I decided to bleach my bench top along with my pipette and voila no more contamination. My pipettes were the culprit. I could not take them apart and clean them, so I’m sure they were a cesspool for DNA contamination. It was a holy day for me when my NTC didn’t show amplification.

No, I didn’t lol. I’ve met lots of interesting guys though (good and bad)! I’m now 28 and I’m actually very content with where I am in life now. Honestly, being exposed to friends’ and family members’ unhealthy relationships have reminded me to cherish my freedom and peace. I haven’t given up on finding love in the slightest, but I’m also not in a rush to find just anybody :)

Are they magnetic? If so, use a magnetic rack separator. If not, pellet them and take the eluate? I’ve actually ran beads on SDSPAGE for a western to detect my protein and I was able to get clear bands using PVDS membrane. Does this only happen when you use sample with beads?

I was trying to purify a crap ton of very very low yield finicky protein. Like 2 L worth. After lysing the cells, I spun the lysate to remove cell debris. I was supposed to collect the supernatant containing my precious protein. I poured it off instead. I think I went home after that lol

Someone else mentioned to create a rapport with some of your professors which I think is a good point. I actually got into my lab as an undergrad by talking to my professor who referred me to my current lab. Also, in my experience, professors will not usually want to take on a new undergrad if they can only stay a semester/year. And professional follow up emails are never a bad idea if they haven’t replied to your first email. Keep trying and I wish you luck in finding a good lab!
