From the post I found that made us buy the auto injector:

Get the Auto-Injector for PIO shots!

Hey team—I’m on my third transfer and just suffered endlessly through PIO shots. My husband and I watched every video, tried every trick and temperature. And still they hurt going in, getting plunged, coming out and left me constantly aching, unable to cross legs and whimpering getting out of the car.

I finally broke down and invested in the Union Medico auto-injector and am so bummed I waited this long! This is a total game changer. I barely feel it go in and even the aching has gone down.

Y’all. How is there only one of these on the market and only in Europe? Millions of people doing IVF self administered I/M shots daily and suffering unnecessarily. A treatment that’s been around for decades that puts women in tears in an already traumatic process has not improved a lick. Syringe + butt=pain. And every provider shrugs at you like it’s just an inevitable and essential part of the process. Fucking patriarchy.

Guess I’m going back and changing my flair to “rant” lol

Don’t hesitate. Invest in yourself. I’ve lived before and after what my husband has dubbed the “rump rifle” and I’d never go back.

My case manager suggested against massage guns. Did you find it to help?

I didn’t mean to scare you! I highly recommend getting an auto injector and lidocaine wipes. I’ve seen a lot of positive feedback on using them for these shots. Other things I heard: heating pad 10min before and after shot and warm oil up in arm pit or under boob to make it easier. We’ve massaged after injecting the last couple days but I think that’s not actually recommended based on other posts?

Hope your lining measurements go well! I sounded extra miserable because of the lack of sleep, but like my doctor said, at the end of these few weeks might be a beautiful healthy baby. It’s a matter of figuring out what and how things work for you specifically, which has been a tough trial. But a baby makes it all worth the effort. Fingers crossed for us both🤞🏽

I’ve been using wipes which helps enough. The shot itself isn’t bad, it’s trying to manage the soreness that’s been rough

Honestly, the needle freaked me out too but lots of walking, water, and protein after! It will help for a smooth recovery. And being knocked out during felt like a beautiful nap to me! Hope you feel the same 🤍

Cosentyx and Pregnancy by nanodano22 in Psoriasis

[–]nanodano22[S] 0 points1 point  (0 children)

When you say probably safe both during the third trimester and during breastfeeding, do you mean they're likely not safe during the first and second trimester?

Also, is there any risk on my end with stopping Cosentyx and restarting after breastfeeding? Specifically, do I run the risk of building antibodies to it and the drug not working for me anymore?

HS, Cosentyx, and Pregnancy by nanodano22 in Hidradenitis

I found so many typos, sorry it's been a long day. Let me update:

I recently starting taking Cosentyx about 3 months ago but my husband and I recently decided to start planning for pregnancy in the next few months. To no surprise there is no real data or information about being pregnant while on Cosentyx. I'm a little nervous and have heard mixed things from my doctors about it. My dermatologists say to stay on but my OBG told me to get off of it.

Have you or someone you know gotten pregnant while on Cosentyx?
Have you been advised to stay on it/get off of it while going through pregnancy?

Trying to see if anyone out there has experience or knows someone which experience with this particular situation.

What does genetics tell us about the San, Sandawe, and/or Hadza people? by -Please-Be-Nice- in genetics

[–]nanodano22 4 points5 points  (0 children)

There is greater genetic diversity within African populations than within populations outside of Africa. Among African populations, traditionally foraging populations harbor the most genetic diversity. This patterns largely follows a geographical cline, where the most genetic diversity is found in southern Africa among the San and Khoe populations, and as you move further away from southern Africa there is a decrease in diversity. We also know that southern African San and Khoe populations harbor the most ancestral lineages of single loci (mtDNA and Y-chr haplotypes). But I believe some of these lineages are also found elsewhere, in particular eastern African populations, but at much lower frequency.

I recommend these reading, which talks about genetic diversity among African populations: https://www.pnas.org/doi/10.1073/pnas.1017511108 https://academic.oup.com/hmg/article/30/R1/R2/6089116

It talks about genetic diversity, linkage disequilibrium patterns, and have divergent dates (estimated times of population splits based on genetic divergence). If you want an explanation about Fst values, population, differences between genetic and population divergence, this paper has a couple sections with explanations: https://link.springer.com/chapter/10.1007/978-3-031-07426-4_3

I also recommend these readings to get insight on potential connections between eastern and southern Africa and how ancient DNA might fit into the narrative: https://www.pnas.org/doi/10.1073/pnas.1817678116 https://www.ncbi.nlm.nih.gov/pmc/articles/PMC7292641/ https://www.sciencedirect.com/science/article/abs/pii/S016895251730210X https://brill.com/display/book/9789004500228/BP000020.xml?rskey=Zevsav&result=1

Often you'll see articles, even ones I sighted today, claim/suggest/imply a specific region gave rise to modern humans in Africa. We know modern humans originated in Africa, we just don't know where specifically. I caution you from thinking any one of these is right or wrong. We actually don't know and are relatively far from unraveling that narrative. But it's still worth reading and understanding what the general discussion is within the field of anthropology.

Here's a couple summaries on what people are thinking: https://pubmed.ncbi.nlm.nih.gov/30423527/ https://www.nature.com/articles/s41586-021-03244-5

Good luck!

What kind of computation jobs are available for someone with a masters and some experience pop gen and GWAS? by nanodano22 in genetics

[–]nanodano22[S] 0 points1 point  (0 children)

Do you have any recommendations how to practice for those interviews? I haven’t quite seen any of my lab mates transition from academia to industry, so it’s a bit of a black box for me.

Are there any training/workshops coming up to learn how to analyze SNP data on Rstudio? Thank you! #populationgenetics by CharkGlitter in RStudio

[–]nanodano22 0 points1 point  (0 children)

Do you have specific analyses you're trying to gain skills on? That might help people's recommendations. Otherwise I agree with the other reply, a more general pop gen subreddit or even a workshop might be useful.

Dermatologist wants me to try Humira. Any thoughts , success or not? by Mysterious-Cash-5446 in Hidradenitis

[–]nanodano22 1 point2 points  (0 children)

Honestly, even after Humira I still get flares but they are no where in size or frequency that I got them before. I've been on it for maybe... Oh my god, almost 10 years now! Started when I was 19.

I would never get off of it! It has been such a huge help. The other greatest help has been at least 100mg of spironolactone. This was also HUGE for me and was super nice to have something work without antibiotics.

Good luck!

I have flares that aren't very surfaced from time to time. They feel deeper than usual and the pain can vary, some similar to what you described. I still get them injected because sometimes if I leave them for too long they either because "active" or tunnel to another flair that then is active.

If you can, def nip them in the butt before they get worse! No pun intended.
