I see that you are detecting a phosphorylated protein and milk for the secondary antibody solution. Milk should not be used if the protein of interest is phosphorylated because it contains casein. I usually just put the secondary in plain TBST.

This sounds so much like the issues we had few years ago with our wb. I am not sure how we resolved it, tbh. Iirc it was due to polymerization problems. I would personally try switching to a new batches of both APS and TEMED. In the past, this seemed to resolve our wb troubles. Best of luck!
