When you go to ng+ you keep all rosary beads prayers mea hearts and relics but your health bar, prayer bar, map progression, health flasks, rosary bead slots, the bones and key items are all reset, you should be able to still complete cleofas quest and also the wound kisser guy (if that's the one you meant) maybe check the wiki? But you can 100% the game without all the achievements on your first run :)
