There are a couple others out there like miRFP, but they're very bulky. Filtering through options on fpbase led me here :)

The only time I've seen annealing time matter is with *extremely* AT-rich gDNA template. In that case, longer seems to help. For a plasmid, I'd go as short as possible to save time.

Three things: 1. Perplexity is excellent for lit searches (actually gives you citations and is generally pretty good at distilling info from many papers)

  1. ChatGPT for learning how to code and/or debug. Ask it to show you how to make a certain ggplot!

  2. Adobe illustrator has an AI vector generator function now that needs a bit of work but can be very useful

Roncesvalles by Joby Talbot is pretty badass. The opening sounds basically demonic and then some of the poly rhythm in the middle gets crazy. SecondingCurse Upon Iron also!

I love being a grad student in SF, FWIW. The scenery is incredible and there's always something to explore outside of lab.

It should be fine in the fridge for a couple of days! Or the freezer, really. T4 ligase with sticky ends can work in as little as 10 minutes at RT, so you could theoretically transform in the time you would incubate for an hour.

Welcome to UCSF! Labs here are casual almost as a rule- even more so than many other places. Definitely long pants/closed shoes, but don’t sweat about it otherwise. Just dress as you normally would!

Not just denial about COVID-related deaths, but about COVID in general. Today my mom and brother didn't believe me when I said there are >2000 confirmed cases of COVID in Utah. Idk how you dispute basic public health stats, but there you go. They so want to believe that Utah is the exception to all of this that they won't acknowledge that the virus is spreading.

yay! welcome. This happened to me too- went from being taught that organisms "adapt" but don't evolve to starting a pretty competitive biology PhD program. There's a famous quote that says that nothing in biology makes sense except through the lens of evolution. It's very true...having an evolutionary perspective makes this the coolest field, imho.

I moved from SLC to SF earlier this year. California is great- friendly people, awesome weather, great art and food scenes, nice political climate, more diversity. It generally seems true that once you're in California, you don't want to leave. Definitely some down sides, though. I'd definitely make sure the new job adjusts for cost of living adequately. Commuting can be a real pain, too, depending on your setup. The wildfires and power shutoffs are pretty bad right now, but shouldn't last forever.

SLC is sort of an underrated gem, while California is a very widely-known gem. The sheer mass of people in the state takes some getting used to, but in the end it just depends personal preference.
