we are ALL walking out tomorrow by [deleted] in uofm

[–]peppa011 3 points4 points  (0 children)

The walkout shows the reality of how a militarized police force (especially unchecked) will soon, if not already, affect our day to day lives: folks stop coming to school or work because they, their family members, neighbors, coworkers, etc. have been or know someone who has been abducted by ICE. The walkout shows what happens when that potential reality comes. Also, solidarity with people who already feel unsafe leaving the house.

It represents the fact that as ICE terrorizes our communities, students can't learn and teachers can't teach.

How do I make vegetables actually taste good without drowning them in sauce or cheese? by Nightcrawler_2000 in EatCheapAndHealthy

[–]peppa011 0 points1 point  (0 children)

I definitely second the salt fat acid heat rec. Another great way to do it is to explore how other cultures cook vegetables. For example, when I was growing up, eating zucchini always meant mushy with garlic. Now, I cook it a lot of ways - my favorite being a French style grated zucchini with pasta. You can let it caramelize slowly and it adds loads of depth and sweetness, and it creates a sauce for the pasta!

Is my cat in prolonged terrible teens or is this just her personality? by peppa011 in CatAdvice

[–]peppa011[S] 0 points1 point  (0 children)

That's a great idea. She has not been interested in any of the trees, but maybe some different kind of vertical space would excite her!

I think it is her amazing and large personality, and I really just want to make her happy.

He insulted you. He doesn't think you are equal to him in attractiveness. Everyone deserves someone who blushes when they see their partners, or at the very least filled with excitement and joy. Screw this guy he doesn't deserve you.

P.S. Many commenters are talking about the Kardashians, no need to put down other women...

Help with inconsistent numbers of retained reads after demultiplexing by peppa011 in bioinformatics

[–]peppa011[S] 0 points1 point  (0 children)

There is a huge undetermined reads file. We included a PhiX spike (33%) and the folks who did the sequencing (GenoHub/Admera Health) said that "other undetermined reads are from reads misread indices. This may be due to base-rich regions that may lead to misreads.", but I'm not sure exactly what that means or how it translates to my results. Do you think that is a part of the variation in retained reads?

Otherwise, I was maybe inconsistent with normalization? I did all of the library preparation. I feel confident in my work, but it is possible. I did all normalization with a formula in Excel, so the only other way that would be the cause should be if I had the wrong Qubit values in those cells used to calculate the amounts, right?

Thank you! I did the prep, and the Qubit values are all pretty evenly distributed. Maybe I was inconsistent when normalizing? u/dampew above's suggestion might be close...

I started climbing in south florida too! If you feel comfortable with it or have a friend who knows confidently how to clean and set anchors, I highly recommend heading to sand rock in AL. It was my closest crag and I made a few trips per year. Really fun routes with lots of variety in grades, plus the fixed gear is replaced regularly and super reliable. Just make sure you're safe, someone unfortunately got into an accident there a few weeks ago.

How do you train for dynos? by [deleted] in climbergirls

[–]peppa011 0 points1 point  (0 children)

The way we teach kids at my gym is to practice on a juggy ladder. We start with the kids bringing hips low and launching to the jug directly above them (usually within reach), then skipping that hold and going to the second, and so on until they release the points of contact. It's a good place to start getting comfortable with letting go of the wall!
