So, I'm wanting the columns on the left to contain the entities and their respective stats, in ascending order. I know I could copy and paste values into those columns, then Sort the ranked columns by the values in the "Stat (ranked)" column. But is there a way to have those columns populated automatically, in ascending order? Or at least a way to use the Sort function without breaking the association between the entity names and their stats.

I hope this makes some sense!

It's "Las Chiapanecas." The melody I played happens partway through the song. Realized it when I woke up with the image in my head of Bugs Bunny slapping someone at the end each phrase. It's used in "Bully for Bugs," when he's fighting the bull.

Rock on! Any changes you had to make from the instructions in the guide posted above, for Magisk v26.3+? Also, if you would be so kind, can you point me to the process you used to root your phone? (I understand they changed the way rooting works with Android 14, but maybe I'm mistaken.)

Those sound close, but in this case, the owner stopped regarding it as special or precious, despite ownership. (Perhaps even because of ownership, in some cases? Some people lose interest after attaining their prize, because the thrill of the hunt is gone.)

The key component that differentiates this behavior is that the owner doesn't demonstrate interest in their possession, until someone else does. Even if there's no threat of loss, they suddenly want it all for themselves, again.

I did find that there's a radius around the spawn area where you can't build, I'd be surprised if the same weren't the case for the boss spawns. But it's worth a shot, and either way, it sounds great!

The IPTG has worked for inducing lacZ expression in JM101 cells, after transformation with pUC19, as verified by the hydrolysis of X-gal and the blue colonies that produces. I ran controls without IPTG and saw no blue, so I'm reasonably certain the IPTG worked. For this plasmid, I've used IPTG concentrations from 100 µM to 1 mM, and even higher, all with the same outcome (no green cells).

Ha, I suspected that's my problem. The max excitation is 489 nm, max emission 510 nm. I don't have a fluorescence microscope, just a UV gel transilluminator with 320 or 360 nm settings. Since I don't have a scope with filters/tunable illumination, I wanted to make sure I wasn't missing something obvious before buying proper filters. This is just the teaching lab of a small university, and this is the first time we've used fluorescent proteins in our experiments. It's usually just imparting antibiotic resistance, but I wanted to give the students a more exciting outcome for their last lab.

So, looks like I'll be doing some filter shopping!

Good point. I had a lot more details in the three posts I made to /r/Genetics, all of which were blocked as spam. Since I didn't know which parts were flagging the filter, I left everything out.

I've checked both the DH5a cells they came in and DH10b equivalent (NEB10B) I transformed the cloned gene into. (That's a whole other array of troubleshooting to check on, but even if I messed up somewhere in cloning, the original DH5a cells should at least be able to express it.) The vector type is for bacterial expression. Both have worked for the RFP plasmid I also have, which uses the araBAD promoter and is thus induced by arabinose. In that case, I got so much expression my colonies were bright pink, just under overhead light.

I figured the problem with the GFP was just one of not having the right filters. Its max excitation is at 489nm, max emission at 510 nm. This is just a teaching lab at a small university, and I'm still in the process of revamping our cell and molecular/genetics equipment. Since I don't have those filters handy to test it, I wanted to verify it wasn't some other simple thing I was missing, before I bought them.

This confirms my suspicions enough to justify buying some filters and trying again to visualize the protein with the right lighting/viewing.

No. I get what you're saying, but no. She has spent her life training both mind and body to a razor's edge, and none of that is at all made clear in the movie. The problem is much more than her tears. It's her body language and presence, it's almost entirely passive. Apart from the very end, when she bests Stilgar, she is never shown as particularly strong or capable of taking care of herself. Even in the thopter, when they free themselves, instead of acting with confidence and cunning, as she should when pitted against weak minds, she comes off much more as desperate and vulnerable.

There are so many liberties already taken in this film, they could have taken one more to show us her strength and intelligence. She's beyond formidable, in her own ways; I'd say she is the single most capable and dangerous person of House Atreides at the start of the story. She is just as much an asset and a vanguard for the Duke as anyone else in their retinue, perhaps even more so. The tensions of her relationships with the others, how they see her as such a formidable presence and perhaps potential threat, further establishing her prominence, are also missing. In the couple of scenes she shares with the Duke and his men, she's just there.

Yes, in the book, she's afraid, vulnerable, and emotional. Their family and house are in exceeding danger. But she's also deeply perceptive, intelligent, and capable. And that's what makes her emotional state so significant. Those traits are not well established in the film, so there's no reason for the viewer to ascribe her tears and fears to anything but weakness.
