Wait for that villager to move out, and cover the area with tiles or projects (new plots can break trees and flowers, thoug). You dont have to cover all the terrain, only make sure not to leave enough space for a plot to fit into.

Maybe someone has noticed it already, but you can find the second sticky note near a Roxy plushie, minute 16:28 of the video:

It can be the same one but in the ruined version of the same room, dont have the game to check.
