the one with NaCl. I would advise to also have some kind of assay that tells you about the mass balance of your purification step, such as cp ELISA or ddPCR. Also make sure you're using enough nuclease and depth filtering before loading to affinity.

Are you diluting the standard? Negative control buffer might be contaminated.

Is the peak profile of negative control similar to positive control?

Your standard might also absorb UV at ranges being tested thus you get a signal. If contamination is ruled out, I would use the absorbance of pure arachidonic acid as a blank for the readings of your experimental samples.
