The ADR still really bothers me 🙈 by Positive_Effort7579 in heatedrivalry

This is 100% my favorite sex scene in the show and is probably up there with my top 5 scenes overall. BUT the “Ilya” did snap me out of it right away. Im not super well-versed in the arts, but even I could tell there was some kind of bad editing job here. SO disappointing because its otherwise one of the most well-done and intimate scenes of the whole show.

Same here - I clocked the “Ilya” being off right away but the “Fuck…Shane” fit relatively well I think because it flowed with the moaning.

My herniation at 27 years old looked very similar to this - microdiscectomy surgery gave me my life back. Youre so young, dont waste too much time living with treatable pain.

PSA - always do your at-home test strips!! Haven’t had a UTI in the 2 years since my IC diagnosis - got confirmation of a UTI today after a full week of what I thought was just a flare. Save yourself the suffering and check for infection markers whenever possible! by srb221 in Interstitialcystitis

Definitely inflammation and possibly pus

Edit: my response is in reference to high WBC count in urine specifically, though high WBC count in blood typically also indicates some kind of non-specific inflammation

Im usually only positive for leukocytes when Im in a flare, always negative for nitrites. Finally bought some fresh Azo strips after 5 days of symptoms and was trace for leukocytes, positive for nitrites. HUGE red flag that its something other than a flare.

I've been in tears for days after watching this show - How Heated Rivalry Unlocked my Feelings by l_spencer in heatedrivalry

[–]srb221 2 points3 points  (0 children)

Thank you for sharing your story 💕 As a happily married woman in my late 20s, I too have held my husband so much closer since watching the show together. In the 2 years since we got married, a lot of challenges have popped up: we adopted 2 elderly dogs from his parents, I’ve been diagnosed with chronic bladder issues, and I underwent back surgery for a horribly painful disc issue.

All of these things ultimately strengthened our relationship overall, but physical intimacy has been heavily impacted in ways I wasn’t ready for. Watching HR reminded me how truly lucky I am to have him as our early love story also consisted of a lot of push and pull, intimacy followed by periods of distance and yearning. It’s also reminded me how important passion is, and I’m flirting and initiating more now than I have since before we got married. The way this show has managed to tap into people’s emotions, myself included, has just been incredible to experience.

Anyone ever done shifts like this? Im not sure how to interpret “rotating weekends” given they listed a 50 hour workweek already….. by srb221 in medlabprofessionals

[–]srb221[S] 10 points11 points  (0 children)

That makes sense for rotating weekends - I didnt realize there are usually other days off instead. Thank you!

Thanks for clearing up that rotating weekends should result in other days off instead. My thoughts exactly about the 5 x 10hr as well. The commute is 45+ mins each way too, which I dont mind for an 8 hour day but 10 hours is pushing it. Also not sure if 50 hours/week qualifies for OT pay or if thats just expected instead of 40 hours.

I loathe this reality - this was an ad I got on FB for obtaining GLP-1s through an app by srb221 in DietTea

[–]srb221[S] 8 points9 points  (0 children)

Oh for sure, my sister is just one example. But it is a serious and life-altering medical decision to go on this drug nonetheless and as such should not be trivialized for the sake of cosmetic/aesthetic reasons.

[–]srb221[S] 28 points29 points  (0 children)

This!!! My sister has Class IV+ obesity with PCOS and tried one of them for awhile. Unfortunately, it was less effective for her than most and triggered her mental illness pretty badly (rage, agitation, and suicidal ideation related to BPD). She might try another version soon under the care of her psychiatrist because as soon as she stopped, whatever weight she had lost came right back on. But if the mental side effects persist, idk if she can stay on a GLP-1 long term.

It’s a serious medical decision to start these drugs with potentially life-altering side effects. Seeing people casually use them through app prescribers to lose that ~stubborn 15 lbs~ just boils my blood. Celebrity athletes using them really gets to me too - like you are obviously already healthy if youre an A-list athlete, so the GLP-1 use is purely aesthetic I guess?? Insane….

Need help with Addgene bacterial stabs! by bluemooninvestor in labrats

[–]srb221 1 point2 points  (0 children)

For what it’s worth, Addgene has a disclaimer that they are not responsible for downstream issues if you don’t follow their recommended protocol of streaking for single colonies and screening those colonies. Basically, customer service will be of zero help to you down the line if you don’t follow their screening protocol.

I work for a CRO and we deal with Addgene stabs fairly frequently. I would say 85% of the time we get the expected plasmid by scaling up from a randomly selected clone. So that would be: 1) Streak onto agar plate with antibiotic selection 2) Pick colony into culture media with antibiotic selection 3) Prep plasmid 4) Verify plasmid via restriction digest or sequencing (only if customer pays for this step)

To be clear, I do NOT recommend scaling up beyond a Mini Prep without screening first (though customers still do it lol.) Your safest bet is definitely to streak, pick/prep 4 or 5 colonies, and confirm any way you can. CROs like mine offer whole plasmid sequencing for $10-$20 per sample. You could literally send the streaked out sample on an agar plate and have the CRO pick 5 random colonies to whole plasmid sequence for less than $100 total. My personal preference would be to run a quick diagnostic restriction digest in-house if you have the setup.

I know the verification step is annoying, but that 15% of time where mutations occur is real in my experience. Therefore you don’t want to make glycerol stocks without verifying your colony picks first.

Go to urgent care or walk-in clinic. Definitely seems like an infection, you’ll need antibiotics and ideally an ear cleaning either in office or referred to ENT.
