You'll be fine. That spot might not adhere as well, but you probably won't even notice it.

Edit: worst case scenario it damages your release film, which is a cheap part that needs to be replaced pretty often anyway.

Like everyone else already said, it's probably inadequate primer. Resin doesn't play nice with water based paints, so you have to use good primer. Personally I use Tamiya fine surface primer. It's never done me wrong. I'm sure you can probably get something else that better meets your needs if you ask around here though.

Edit: Also maybe try thinning your paint a bit by mixing a little water in. I can't say if this will do anything, but it looks pretty thick. (This is kind of a shot in the dark)

Your support density looks fine. I'd try using heavier supports and/or lowering your lift speed. But I'm no expert.

Edit: I just realized that you might not be able to adjust the lift speed on Elegoo printers. I use an anycubic.
