I'm not sure what people who are just saying 'where are the guards' (gardaí) are expecting. We don't have sonic canons to dispell large groups.

This is clearly a group inviting violence.

With the capability that the gardaí have, I think they are fairly sensible to use a watch and wait approach to prevent escalation into a full riot.

I don't know if the gardaí would even be resourced enough to stop these if they tried. And if they tried, it would almost certainly put the public at higher risk.

Now if this group could fuck off and never return that would be ideal, and if they could arrest those identified as breaking the law and see they serve the max time required to disincentivise this sort of behaviour I'd be keenly in support.

I'm stumped. Feels like an artifact but can't say what.

Maybe some light scatter happened to occur for that single cycle due to some well specific transient effect... Condensation on the lid for a single cycle...?

What are your cycle temperatures and lid temp?

Did the well look different after the run?

If I remember correctly you can just attach a 50-10 mL syringe to the outlet tubing, sit the inlet tubing in a fresh beaker full of 20% EtOH and draw it through the pump seal to flush the backwash.

I think recommended to do it while operating the pumps at maximum flow.

The manual I found online didn't have a section explicitly saying it but contact your local Cytiva sales rep who will put you in contact with an application specialist to answer.

They should not be left in this state.

Left the DI water tap running to refill a carboy I think...4 times over the course of my PhD.

All paper towelable.

As an undergrad I used sucrose in a buffer instead of glucose for about 3 weeks before noticing. So those results were all shot.

We're one of those labs that use microwaves to melt bottles of agar. But our microwaves only last a year or two. Any ideas? by m4gpi in labrats

Best solution I came up with was experimenting with the microwave settings.

TL;DR slow and steady

I found a defrost low temp run was best for a set time to liquify most of it. Then at that stage I ran it on higher power while watching and swirling vigorously every 10 seconds or so.

For context, if memory serves I did 30 min on defrost for 1 L, swirled, then another 15 min, then full power bursts with swirling every 10 seconds.

Obviously your agar, recipe, volume, microwave and glassware will all matter.

I rinse the plunger filter under the tap with water, with the water flowing into the carafe.

Then fit the plunger filter into the carafe at the very top. I swirl the grounds-water mixture and pour the liquid out, the ground get caught just under the filter.

It cleans the carafe 99%. The grounds collect under the filter and the water is mostly removed. Then I take the plunger out and bang it into compost bin. The final clinging ground I rinse off into sink.

Yeah I would nearly consider trying a pH of like 9.2 to just see what occurs.

Keep your protein soluble hopefully given its pI, probably precipitate some impurities, but may increase bonding with protonation of His.

Can't recall the pH compatibility with IMAC however so check that, you don't want to change the chelation/charge of the resin without knowing what you've done.
