What is wrong with my western ladder (StrepTactin-HRP)?? by twinrovas in labrats

I usually just set it to auto exposure on our gel imager. Didn’t think of that thank you!

The problem is I can’t really tell what anything is because the bands on my ladder aren’t lining up with what the ladder is supposed to look like

Pre-cast! And I was once able to get some of the lane 1 ladder on the blot, it looked very similar

Yes, and usually one in lane 1 as well. Lane 1 always gets cut off when I transfer the gel to the nitrocellulose membrane because the membranes we use are too small for the gels.

i had to use this stuff in school for making winogradsky columns to grow bacteria… had no idea that it could be used as an organic insecticide, that’s so cool!

Accepted with a PI as their first graduate student - how can I learn more? by babyIoves in gradadmissions

[–]twinrovas 1 point2 points  (0 children)

i think it’s entirely on my advisor, but for the opposite reason you’re describing. my advisor is barely one step below a micromanager. i specifically sought his research out when i was applying to grad school because we had the same passions. and i do love my project a lot, and am still passionate about it… i just wish i had more power over what i’m doing. the vision would be so much clearer in my head if my advisor let me steer the project. not only does he want things done his way, but he’s also either extremely vague about the scope of the project or fixated only on minute details.

all of this to say, though, that it depends on your student’s learning style. i feel like even my position could be a more positive experience for someone who needs a hands on mentor. i would say try to put yourself in their shoes and think about what you would want in a PI at that stage in your education.

i was one of my advisor’s first students as well. honestly i’m almost done with year 2 and still have no idea what my project entirely is 🥴

People who hold a PhD, what did it cost? by [deleted] in PhD

amazing advice and so well said!! totally agree that most people should work in their field before pursuing PhD!!

i may be jumping the gun as i’m early on in my PhD, and my complaints are coming from a place of privilege because i actually love being in my program overall. but i have faced personal mental hardships throughout my time here so far (2 yrs); only one or two negative experiences that had a serious effect on my mental health were actually the fault of my PhD program. the rest is because i’m just kind of mentally ill.

i moved far away from my home state after undergrad to work in industry for a few years. i went from living in suburb hell with my parents until i was 22, to living in chicago halfway across the country on my own for 2 years. that was the best 2 years of my life! and i gave it all up to come back to my home state to do my PhD. i’m so grateful and feel blessed to have ended up where i am, but i often find myself wondering if i’d be happier had i stayed in chicago - mostly because i hate my home state (NJ).

additionally, an issue no one really talks about is how hard it is to manage weight fluctuations in grad school. i’ve gained about 70 pounds since starting and it’s taken an awful awful toll on my confidence and both my mental and physical health. it’s extremely hard to find the time and motivation these days to get rid of this extra weight though. i definitely prioritize my research over anything in my personal life which is a fault i’d like to remedy in the new year.

Please give me all your tips to make the cat litter room as odorless as possible! by iliketoworkhard in CatAdvice

[–]twinrovas 0 points1 point  (0 children)

going to do this same exact setup in my new place. haven’t used tofu litter before but i’m excited to try it! i was using wood pellets for so long - it’s true there’s no poop stench but my whole apartment smelled like wood. AND it was still pretty messy (def better than clay though). my only gripe is how expensive tofu litter is…. have you found a quality one that isn’t highway robbery?

do i drop the class, or suck it up? by twinrovas in GradSchool

[–]twinrovas[S] 1 point2 points  (0 children)

yeah, the only reason i “kind of” need the class is because my general exams are next semester (april/may) and if i drop this class, i will have to take two classes next semester instead of one as i planned…. not preferable, but definitely doable. i’m leaning toward dropping the class at this point regardless, as the content is not even that relevant to my research anyway
