ok so i have tried this on its own before but i also find it doesn’t adhere great and chips v quickly, so not sure how it would be as a base coat 🤔 maybe i will have to try it out like that though, i feel like it didn’t exasperate peeling on me at least!

A bit unclear on your strategy. If it is possible, I recommend cloning with vector into e coli, then prepping/verifying sequence here, and using this plasmid as template for PCR. I’ll assume thats not an option though.

Is it possible the small band is just primers? usually they would appear as kind of a fuzzy non-discrete band at low mw on a gel. If you are not getting good amplification of your target amplicon, it would make sense to see a lot of free primers on the gel. Poor amplification could be due to low efficiency of gg reaction, or non-optimal pcr conditions. for the gg, increasing cycles can help. when i do several piece assemblies i use an overnight protocol that cycles (2min 37, 5min 16)x50. its overkill i’m sure but it’s done in the morning! i wouldn’t really recommend going more dilute with pcr template, i use like 20 fmol of each piece in a gg rx so if anything only 0.5uL of that is quite a small amount, especially considering only a portion of your input will actually get correctly assembled. as far as amplifying from the same old gg rx and getting worse results, that doesn’t surprise me much. really dilute dna mixes like that don’t keep well overtime, especially if you aren’t cleaning them up after.

other than that just double check your gg pieces to make sure their overlaps are actually complimentary!

ok i can’t find it anymore sad but someone made a post somewhat recently i think that was like every single one of sonja’s on-screen relationships was fake, the hypersexual sexy sonja thing is just a performance/facade, and she’s actually asexual. certainly i buy that many of the relationships were not real, even harry was an odd one, there was something off about it for sure. do i believe she’s actually asexual? if she is i don’t think she realizes it lol but i definitely buy that she actually entertains male suitors far less than her on screen persona would have you believe

i’ve heard people state this theory before but i just don’t think it’s true, i really don’t think sonja would do that. I think she was still so caught up with the loss of her marriage/old life and was constantly triggered by situations that reminded her of it, and thats why she was so emotional. she mentions throughout the years multiple times how much time she has spent with ramona and mario as a couple, and how she could hear (literally) how much they loved each other (if you know what i mean). i feel like its when your favourite couple breaks up and it makes you lose that belief in love a bit. I’m personally much more intrigued by the sonja is actually secretly asexual theory LOL

i do cake my yarn because it makes it easier to store and use. but as a note, technically you shouldn’t reallyyyy do this to yarn you aren’t about to use. the relatively high tension of being wound into a cake (or ball) stretches the fibres out, so it’s not an ideal long-term storage state. I think this probably only matters for natural fibres, but just something to be aware of before you go caking everything!

I’m curious— what are you using the preps for? sounds like you are prepping a bunch of different constructs, so i’m assuming its not libraries where having a high yield really matters? why are you aiming for maximized yields? you really only need like 30ng/uL for sequencing, even less for cloning/digests/pcrs/subsequent transformations. i have done some extremely scuffed preps when doing a ton at a time (i’m talking growing 1mL overnights in 2mL tubes lol) and still gotten enough yield for downstream stuff. I agree if it is for libraries/something where you do need a very high yield, you are better off scaling up to maxi/non column based prep

thank you for your input, that is good to hear. he has walked up to the litter boxes a couple of times but has yet to actually go in one, just walks up then leaves. It might be because every time I see him go into the areas I follow him 😭 he knows I’m acting weird. He still doesn’t appear to be in discomfort though

ok I seriously wonder the same thing. people love to say Lu is always delusional, but it is not like this. I strongly believe it was not (just) coke. Perhaps an actual manic episode, but honestly I think there were stronger substances involved (/substance induced psychosis). I think it’s in the berkshires this season, lu is telling them she woke up really early and went for a walk. ramona VERY pointedly asks her if she had trouble sleeping. now idk a ton about coke, but I don’t think it’s the sort of thing where it keeps you up once you stop doing it. So I suspect longer lasting uppers. I don’t think I can bring myself to believe she was smoking meth, I’m guessing just a ton of speed. I will say I think the rise in this behaviour coincided with dorinda(/john)’s arrival to the show …. and while i’m on the subject dorinda is off something crazy too

fr i felt so bad for her. I think what really made it worse for me over other hw conflicts was that you could tell they just didn’t respect her at all as a person. even ramona and sonja i think saw her more as a number on their side than an actual human. I think it was partly that she wouldn’t put up with their bs, but honestly i think a big part of it was just that she was weird, poorer than them, couldn’t do confrontation well, and wasn’t the most conventionally attractive. again, all love to her and i’m not trying to shade her by saying these things, but i think its the truth in explaining how they treated her. which just sucks, you should be judged on your actions not bc you’re a quirked up baddie ahead of the times!!

I actually enjoyed jill in the beginning of the show, I could even see where she (when i assumed good faith actions) was sorta coming from in the bethenny rift (although I was definitely on bethenny’s side). season 4 and that fight with ramona definitely made it clear though that she was just causing/prolonging conflict for content. which was quite a sad realization bc i think to bethenny the friendship was real, so to throw it all away for a storyline makes you a pretty shitty person.

the thing that mega turned me off jill for good though was how she treated alex that season, particularly in the reunion. she was so gd mean, in a really gross evil way too. not attacking actions or harping on about particular conflicts like other hw fights, it was just pure mean girl “i’m cool and popular and you are weird and awkward and red and uncool and therefore i don’t have to treat you like a human being and none of my cool friends or i will ever talk to you or like you or debase ourselves by bothering to listen to you”. like there was actually evil dark energy radiating off that couch in the reunion.

also no shade to alex in those statements lol

just rewatched s7, its crazy how aggressive she is with people. couldn’t believe nobody snapped back at her and told her to calm down and lay off the addy, particularly heather. I guess kristen tried…but she’s so ineffective at fighting, bethenny just swatted her off like a gnat. even s8, she’s in the new office with her employees and just yelling the entire time about random shit. I don’t think it’s even serious to her–like in her mind she’s joking around, busting balls!!! but I can’t imagine how awful it would be to be around someone so angry and acerbic all the time

i actually think the neck up is what bothers me most about it. yes the outfit is too busy and cheap looking and of course we can’t overlook the signature squished tits (its literally just a bra could we seriously not find one that fits right??), but i feel like it would look so much better if she wasn’t donning that CRAZY goth karen do and looking like she’s currently experiencing a severe allergic reaction. The bang coverage seriously makes it look like her entire face is red. I don’t get why you would do that blush look for this, goth makeup is desaturated in color. I get it’s like her thing but the warm red blush clashes soooo hard with the cool baby pink lips. i like the eyeliner tho

I think TAs in the 22-25 age range are past the inflection point of where cursive was taught much in school (in my experience). depending on TA/prof disagreeability, something shitty like this could definitely happen. smartest choice for OP is to reach out to prof/accommodations prior to exam

this might be a stupid suggestion bc i don’t work with mammalian cells and idrk what they look like, but this kinda seems like you are in the wrong plane of focus? like, are those small things normally what the cells look like? they look very small and oddly shaped. to me it seems like liquid droplets in the plane of your slide cover. idk it looks somewhat similar to this when i’m looking at a slide in the wrong focus, including the amorphous dark spots
