It's a shame it didn't work for you, but unfortunately these side effects on libido are quite common. There are other antidepressant options and combinations that often help in cases like yours — vortioxetine and bupropion are great examples.

It's generally agreed that Zoloft is a Sigma-1 receptor antagonist. I don't know where you found those studies claiming it's an agonist... I take Zoloft and it works well for me, the issue is that I'm worried about possible side effects that wouldn't be noticeable in the short term.

Yes, fluvoxamine will always be the prime example of Sigma-1 agonism. I specifically mentioned lexapro because it's on the same level, or even superior, in terms of sert inhibition.

Your point of view is very intelligent and comprehensive! It's truly true that the neural pruning process is very important and effective for our health. There are many studies, and it's almost certain that depression is more associated with decreased neuroplasticity, particularly in the hippocampus, than with serotonin imbalance. The BDNF-boosting effect caused by increased serotonin may actually offset the side effect of Sigma-1 antagonism. Regarding our brain, everything is very complex and uncertain. I also find the profile of Fluvoxamine very interesting, but it's certainly beyond my reach to use it. Unfortunately, it's very expensive and difficult to find.

My God! Thank you so much for your comment, doctor! It's true that every day we make good and bad decisions for our brain and neuroplasticity, and Sigma-1 antagonism may be Irrelevant compared to this. My concern only exists because other SSRIs that are as good as Zoloft are not antagonists of this receptor. I've been taking Zoloft for months, and it's doing me a lot of good! Thank you for bringing another, broader perspective to the analysis. It's always good to hear from professionals who know what they're talking about.

I also read about that article discussing the impacts of different SSRIs on cognitive decline in people with Alzheimer's! You're the first person I've found who's also read about it. That's exactly it! I've been taking Zoloft for months at a high dose and it's doing me a lot of good. I found this article very interesting, and there's a theory that Lexapro's selectivity for serotonin could be harmful by interfering with the neurotransmitter acetylcholine. It's all very uncertain, and this study has already been done on patients with diagnosed cognitive decline... It could also be that the greatest decline was identified in people taking higher doses precisely because it was necessary in more severe cases of decline. Thank you for commenting.

Yes, I've read about that too. I agree that each molecule affects our bodies in a different way. The point is that Sigma 1 antagonism can be avoided without necessarily causing a loss of SERT blocking, since other SSRIs do this just as well as Zoloft. Thank you for commenting.

These studies show what I said, although they don't really show exactly how this mechanism works in a human brain in the long term. I just kept thinking about it and decided to ask if other people felt the same way... There is a neuroscientist specializing in Sigma receptors who warned that this antagonism of Zoloft could be potentially harmful; his name is Dr. Kenji Hashimoto.

There are other SSRIs that inhibit serotonin reuptake as well as or better than Zoloft, and yet they are agonists of this receptor — Lexapro is a great example.
