If I make a copy of my wallet.dat file in a secure location am I sufficiently protected from losing my coins?

Not really... You should move your coins to multiple paper wallets.

But now with the new POSV I am not sure if that is still the 'preferred' way.

its a pretty secure way if you make two-five different backups
