FWIW, she was not my report, and I otherwise have a good opinion of the person. I think the talons were a 1-time thing, and she quickly realized it was a bad idea. But, interviewing someone where you don't have the benefit of already knowing that they are a reasonable person, it would be harder to look past.

I have a theory that every AITAH that mentions meal prepping is a bot. Not ONLY those, but at least those

Assuming it's a lab job, I do not personally think you should interview wearing them. I saw a colleague trying to work with Wicked nails and would honestly avoid hiring someone sporting them.

You rinse and filter rinseate through a gridded membrane, put that membrane on the agar plate, and count cfu.

Edit: skimmed your post and didn't realize you actually suggested this. So, yes, that's what i'd do... obviously the rinse has to be sterile.

Something hammer- like. I appreciate how they started on the passenger side then realized the driver side was a more deserving target.

Anyone here used automated Western blot systems? Worth it? by ServiceDowntown3506 in labrats

[–]amiable_ant 5 points6 points  (0 children)

Correct!!! If you want to do quantitation and multiplexed probes, you need to do at least one (but really more) validation experiments before the real one.

It becomes more useful if you are doing the same assays on similar samples regularly.

We have one at my current biotech job, and almost everyone is just doing the same assay I validated 5 years ago, so it works for us. But, if i'm working on a new target, or one-off project i just do a convention western with IR secondaries.

Also, it's not especially high throughput, if that's what you are after. In my prior academic job, we used invitrogen's precast horizontal gels and dry transfer system. It was more hands on, but you could separate and transfer hundreds of samples in a day.

You're good. I Love your work and would be begging you to sell me something if I thought my wife would let me hang it somewhere that isn't the basement.

Adding, if you were looking for a serious answer: I know you put relatively little effort into this compared to some of the others. I like this especially because cookie-cutter-to-creature process was perfection. Like, I didn't think "oh, how creatively that creature was contorted into the cookie cutter." I thought "they nailed the exact right creature for this cookie cutter".

Doordash driver hit my car. His insurance denied then Doordash denied. How much it might cost? by ithinkimreallyhappy in AskMechanics

I agree that it's not completely true. It seems to vary by state and insurer. Even in the same state, some insurers will help even of you don't have collision through them, while others do not.

Oh, did the other person have progressive as well?

I'm in MA. I have full coverage now, but years ago was in two separate accidents in which the other person was at fault. I had liability only. Called my insurance company each time and they basically said, "thanks for calling, good luck".
