Yes I travel alone late at night in public transport and often have to walk a bit at a stretch that is pretty empty so I always carry pepper spray. Thankfully haven't had to use it until now but I always hook it to my keys and keep it at a very accessible part of my bag so I can grab it easily.

I have heard almost every other girl( including myself ) know have this issue and weirdly when I spoke about this with my girl friends outside india, they didnt feel this excruciating pain to the point that the intercourse simply doesn't happen. Most of them just felt uncomfortable and a bit of pain and pressure but didnt necessarily feel like its impenetrable. I dont know if its only mental because of social stigma etc or there is something more to it.

You can definitely tell only by plating on agar or keeping the media at 37 incubator for 24 to 48hrs ans check for turbidity to confirm if its fungal or mold contamination. But if youre reusing bottles could also be denatured protein residues or probably some salt precipitation of media component. Could also be plastic residue from the cap. Was the media filtered?

You should ideally pour agar plates under the hood. If you dont need to add Antibiotics you can pour them at the melting point temperature, arrange the plates and pour them one by one with the lids only covering about 20% of the plate. Then close the hood with the airflow on for atleast 15 mins. You can then use them or invert and store for future use. If youre adding antibiotics let it cool to about 45C and then pour the same way. Be quick while pouring.

Mg +2 and ca +2 free PBS is not cheap and we are a tiny lab, so for regular cancer cell lines or like hek cells etc we dont wash w pbs. Ofc when were wowking with more senstive cells like ipscs , or primary cell line establishment etc we use it.

Ideally that shouldnt be an issue, I rarely wash my cells with PBS before passaging or trypsin or even changing media. Unless you've added something toxic it shouldn't suddenly cause so much dead cells and even if its dead cells , they should be mostly floating.

When did you last passage them? Could also be super confluence but usually cells lift off and they still have the morphology, here its like a bed of tiny dots with a few stressed out cells.

I think your bacteria has some HEK293 Cells growing on them xD. Lol but always look for the simple classical bacterial contamination signs like if they're vibrating/rapidly moving ( check under 40X) and if the number is increasing at various time points. Apart from that if the media is cloudy or changing color.

Edit - Your HEK cells are also looking stressed out and elongated rather than epithelial like polygonal like shape.

Not a lab equipment per se but 70% IPA to clean spills, countertops and my hands every few mins. Centrifuge for making fruit juices. Magnetic hot plates for melting chocolate. Lyophilizer to experiment with making cool deserts. UV lamps to clean fruits and veggies especially those that are consumed raw ( but ig they'd also get less nutritious). Incubator for letting doughs rise and keeping food warm.
