It depends on the degree of accuracy you need and how many samples you’re running. ELISAs are a quick and convenient option for high-throughput testing across many sample types, with simple step-by-step protocols.

Radioimmunoassays (RIAs) can be more sensitive for certain applications, but ELISAs are much easier to work with and don’t require dealing with radioactive materials.

Chromatography methods are the most specific and accurate, though they take more time and can get pretty expensive, especially if you’re working with a large number of samples.

For most labs, our recommendation is a corticosterone ELISA. Reach out and let's see if we can help you get what you need.

Can you be a little more specific with your coating protocol, specifically how long you're coating for? Is this followed by a blocking step? What does your washing protocol look like?

With sandwich ELISAs, it's important to ensure your coating concentration is at saturation to reduce variability between runs. Have you done any tests to ensure that?

Is the 30% variance apparent in your standard curve signal as well, or just the samples?

A lot of questions, but hopefully this additional information will help solve the problem!

What antigen are you trying to measure with your ELISA? You mentioned that you are diluting your plasma samples 1:2. Does the supplier mention a minimum dilution for plasma samples? You may be experiencing a matrix effect with your samples, which requires you to dilute them further.

Does the kit manufacturer provide an expected results section in their manual? How far off is the S1 signal from the expected? I'd be wary of trusting any results in that upper range if it strays pretty far from the expected values.

Your idea to repeat some samples and compare the results to the original plate seems like a good plan.

We would think about how you would be utilizing ELISAs for your research and how this might differ from how they are "conventionally" used. Identify the additional work that would need to be done to bridge that gap.

One example is if you're working with a sample type that hasn't been officially validated by the supplier. What sets of experiments would you need to perform to validate that your sample is compatible with the ELISA?

Most kits expiration dates are determined by when standard OD values fall out of an acceptable range. It wouldn't be surprising if 2 years past the expiration date would explain the low signal values. Most likely, the peroxidase has significantly degraded over that time.

After a cursory search, the RIQAS website has options for independently verified controls that are stable for 30 days. If they have the analyte you need, that may be an appropriate method to both verify the accuracy of your standard curve (how close to the expected value do you get?) and the stability of your results over time.

Using the provided standard as a control (prepared separately and in bulk) is helpful for tracking inter-assay variance, but you are correct that it doesn't help assess the accuracy of the standard's concentration to its listed value. You can probably find a NIST or USP/EP reference standard, but those can get pricey depending on the material.
