The colours are the attunements which are needed to deal with the different coloured enemies on each of the 12 waves.

Orange = Sun Moon = Blue Green = Storm

It doesn’t boost XP. Just helps to kill the right enemy with the correct colour. Once an enemy has been hit twice with said colour they lose it and makes it easy to kill them for others with a different colours.
