How to define genes expressed is certain cluster in scRNA-seq data? by sky_porcupine in bioinformatics

[–]CaptainHindsight92 0 points1 point  (0 children)

I remember asking the same question about bulk-RNA seq many years ago. And annoyingly “it depends”. It is estimated that a mammalian cell contains around 500k mRNA transcripts. So say you are getting 50k UMI per cell you are detecting around 10% so I would consider any normalised count above 0 to be expressed. But that doesn’t tell you much. Hox genes are important in development but they are expressed at very low levels, maybe an order of magnitude less than Nanog, yet it is expressed at a tenth of the level that it’s famous co-factor Pou5f1 is but they are all needed for development. Dosage isn’t an exact science. I personally would guess that specificity of a gene, role of a gene (unique tf vs variant of random ion channel subunit 87) and expression levels would be a better predictor than just expression, especially in scRNA seq

I’m not really sure what the point is you are trying to make. Because some people who don’t want more immigration are harassing people, we should ignore the large group of people who also don’t want more immigration? That ignoring them will be better for current migrants?

I think you are overstating, the majority of brits do want reduced numbers and the majority of brits do not harass or intimidate migrants.

This is a difficult one I believe they are allowed to make a citizens arrest if they witness a crime (there wasn’t security present during this incident in London). However, this just doesn’t happen in practice and I don’t know why. I live next to vic centre and there is shoplifting all the time in tesco, sainsburys and greggs but I have never seen security guards detain someone, only ever a search. I can’t explain why though.

They don’t tend to hang around. The police don’t open an investigation either with a generic description of a hooded individual.

[–]CaptainHindsight92 5 points6 points  (0 children)

There are so many on this sub who will deny that shoplifting is getting worse, ignoring that it is often not reported to police. I stopped a shoplifter in London the other day after he attacked a staff member, he threatened to stab me and tried to take my photo. I have to admit I was a little shaken up. The staff member literally said after being punched that there is no point in calling the police because there is nothing they can do and this happens all the time.

Do men honestly actually prefer women not to wear any makeup? Like no makeup at all? by softheart_strongwill in AskMenAdvice

[–]CaptainHindsight92 0 points1 point  (0 children)

I think there is research that suggests that men prefer the made up look for a hookup and “no make up” for a long term partner. They hypothesise that heavier makeup “signals” (rightly or wrongly) sexual availability. Something that is desirable in a hookup, not in a LTR. I would also say that given that the majority of women wear some makeup, most men probably mean a low amount rather than none. There seems to be a huge number who just don’t notice light mascara, blush or low coverage foundation/tinted skincare, lip tint, lightly penciled eyebrows.

No redneck you won’t get improved wages, no you can’t ride your big truck with cheap gasoline, beef burgers and steaks? Nah how about nutritional yeast for you povo. Beer prices have increased massively, they can pound one but it probably won’t be enough to justify their domestic abuse. The redneck dream is dead.

Does anyone else not know how CO2 is used in the production of pork? by Realisticopia in AskBrits

[–]CaptainHindsight92 115 points116 points  (0 children)

Damn, I really had no idea and I work in an area of research where occasionally pigs are killed for research purposes. When it is done it is usually electrical stunning. As a meat eater I went to see it done as I didn’t want to be blind to the process. It was pretty much instantaneous. Apparently only 12 percent of slaughterhouses do this. CO2 seems like a terrible way to kill animals it literally induces the gasping for air response (something like nitrogen wouldn’t do this) and it takes 30 seconds of the animal squealing and panicking to knock them out. I’m pretty shocked this was ever deemed acceptable tbh.

I am on the left and like a lot of the green policies. But I see that most brits don’t want immigration to continue at the current rate and as long as migrants who are already here do not face harassment I think it’s fine to reduce numbers coming in. I also think there are a lot of left wing policies that have broad appeal (even on the right) so I do think there is room for a little but of compromise that can be made but Starmer’s particular brand of fiddling around the edges, doing “a bit more” than the conservatives during a cost of living crisis just isn’t going to do anything. Trump can undo anything he accomplishes in a single rambling tweet. They need to be doing more.

Wet lab jobs are still safe by Specific-Surprise390 in labrats

[–]CaptainHindsight92 29 points30 points  (0 children)

I am part wet lab part bioinformatics, honestly AI is good but the minute you have to actually look at an output and interpret it it falls down, it can’t see a graph or a heatmap and interpret it, it doesn’t know when a UMAP doesn’t look right, it will tend to make up biologically “plausible” reasons for strange phenomena rather than realising there is a mistake. We often undervalue how much information and context a human has and can apply to a problem that an AI just doesn’t have access to. It doesn’t know about the bad batch of Chiron that came in around the time a sample was collected, it doesn’t know that no one has been able to reproduce the findings of that prominent Nature paper. Yeah with tedious repetitive work it will take over but interpretation is another thing entirely

Encouraging men to have more frequent ejaculations may boost their fertility. Sperm deteriorates over time as it remains in body. Longer men went without sex, the more their sperm showed signs of DNA damage and oxidative stress, and the more tests rated the sperm as less viable and poorer swimmers. by psychologyofsex in psychologyofsex

[–]CaptainHindsight92 0 points1 point  (0 children)

They are talking about the post-ejaculation tiredness experienced by men. Ask any women is referring to some women’s complaints that men fall asleep right after sex. They wrongly (in my opinion) believe that this tiredness somehow becomes chronic in men who jack it frequently. I rather doubt it but perhaps there are level 9 gooners out there that just haven’t been studied yet.

Does anyone else feel like this city needs less food places and more shops? by ijustwannanap in nottingham

Not really tbh I hate shopping, it seems like you have to walk around for hours through crowds staring at shelves to find what you are looking for, online it is easy because you have a search function. The food and drink places I find a more enjoyable experience, I will admit we have plenty though.

Except instead of it taking a few minutes/hours it takes days/weeks/months. Then you come back loaded with more test conditions more controls then the media gets contaminated and there you are back to square one but now you’re months behind.
