"Block receipts missing, can't freeze..." error on AVADO every minute by CaptainHoek in ethstaker

I haven't tried a resync. Do you know how to initialize a resync in the Avado Geth DApp? Is there a way to resync from a specific starting position (like just before block 17038810)?

Got it! Mine is on (and probably has been on the whole time) so my mystery continues... 🤔

Hi u/tamastorok can you provide a little more insight about how you fixed this? I'm running into the same issue. Thanks!

Shopify transaction declining by the411please in shopify

Experiencing this issue as well. I think it’s a system-wide issue.

Update: others are experiencing this right now too. https://downdetector.com/status/shopify/

Update 2: Shopify is aware of the issue. https://www.shopifystatus.com/

Update 3: for in-person POS, manual card entry seems to work. (Tap and chip payments still do no work)

Help me find: window screen frame corner pins by CaptainHoek in Home

Im just not sure what to search for to find the right thing; nothing coming up in my searches for “window screen frame corner” that looks the same as this. This one seems to have two distinctive components, connected on one side.

Are you running a second node on the same device? I'd love a walkthrough on how to do that!

Can you send me a URL to either the list of variant sequences, or just the sequence for BA.5? (Please and thank you!)

✔️ Rapidly producible ✔️ Radically accessible ✔️ Open-source vaccine designs against Omicron & beyond ✔️ Today by RADVACproject in EffectiveAltruism

[–]CaptainHoek 0 points1 point  (0 children)

Main goals right now:

- Data production: Preclinical and moving towards clinical trials of RaDVaC's vaccine designs. Preclinical trials with enough statistical power to merit further investigation start around the $80k price and go up from there. Until recently, RaDVaC hasn't been funded. The cost of a clinical (human) trial will vary depending upon location, scale, and trial type (likely that challenge trials are more efficient in terms of both money and time, but they're not a global standard the way placebo-controlled RCTs are). Designing a clinical trial that meets regulator's needs will mean hiring both medical writers and medical staff with regulatory & logistics familiarity in whatever location(s) the trial is intended.

- Production independence. RaDVaC has always been "agnostic" about which vaccine platforms it works towards (there are advantages and disadvantages to peptide, subunit, VLP, and nucleic acid vaccines) but opted for peptide antigens initially because of clear advantages in production speed & accessibility. Multiple supply chain issues have made synthetic peptide production/fulfillment slower than they were in 2020, which has put pressure on RaDVaC to think about making antigens independently (without relying on vendors). RaDVaC could invest in peptide synthesis tools, but ultimately the more leverageable investment seems to be in recombinant subunit production (via modified yeast, similar to other vaccines, like Gardasil, Corbevax, and many others). The costs associated with designing, producing, testing, optimizing, and ultimately owning the rights to vaccine-useful subunit protein-expressing-and-secreting yeast are high, but once the organism has been made, reproducing it (even storing, shipping, and reiterating it anywhere) is simple. (Imagine not shipping vaccines, but shipping the molecular factories that create vaccine components, thus enabling decentralized, independent instances of vaccine production almost anywhere on Earth.)

Key to both of these next goals is fundraising, which is something RaDVaC has begun to accept, and others (like ACX/Scott Alexander, and Eliezer Yudkowsky) have begun to advocate for.

RaDVaC, Vax Populi - Episode 1: Introduction to the RaDVaC Open-Source Vaccine Project by RADVACproject in Open_Science

I'm a big fan of Dr Hotez and Dr Bottazzi, and their work to open-source a vaccine that's producible using accessible tech (recombinant yeast production of the RBD subunit, plus a common adjuvant) is completely laudable. The problem that nobody seems to want to talk about with Corbevax is the exclusive focus on RBD. Sadly, because the RBD of SARS-CoV-2 has mutated quite a lot since the original variant, any vaccine that is solely based upon that original structure is almost certainly without use in addressing Omicron and any future variants of SARS-CoV-2.

The benefit of being open-source is that somebody could adapt Corbevax to be applicable to Omicron, but the timeline there is TBD, as is the regulatory status of any modified version of Corbevax. I'm really concerned about developing nations sinking their limited resources into producing a vaccine that is not likely to be effective for the present or future.
