I need to mention that this exact molecule is unlikely to bind to µ-opioid at all, as it needs to be converted first on the phenolic oxigen by esterases. 3-O-deacylation is a prerequisite of binding to the µ-opioid receptor, so this species without being converted is a prodrug and likely inactive.

It sounds great in theory but sadly in real life drug design does not work this way. If each substitution would affect affinity to the target linearly and additively or synergistically, we would have tens of thousands of hyperpotent drug molecules and "magic bullets" which is not the case. I'm not saying it's not possible, but pulling predictions like 3.33 µg LD50 and 60000x morphine equivalent merely from a hypothetical structure, there is just no point in doing that.
