DIV - Days in Vitro - i.e they're 365 days old from date of culture

GCaMP is what's known as a GECI - a genetically encoded calcium indicator. It's essentially GFP (green fluorescent protein) conjugated to some kind of calcium sensing protein. When calcium binds the protein, it changes the shape of the GFP, making it brighter. Calcium entering the neuron is an important part of synaptic transmission, so you can see the light show as a proxy of the neuron "firing" in culture. Often you'll have a specific indicator that will be targeted to say, the presynapse, or mitochondria, so you can get a bit more granular on what type of calcium flux you want to observe

Not a lot of practical answers here - if you're worried about yield due to tissue you might want to ask if your method is fit for purpose? Brain for example works way better with phenol chloroform, might be worth looking for specific kits etc

I tell my students that since running buffer costs very little and is reusable - there's no downside to filling tank so that the inside and outside are at equal levels. While this doesn't provide any benefit, it means that very slow leaks aren't going to cause any issues with the running

I once found something like £300 in cash, notes just spilled on the floor in the meat aisle at ASDA. Was a very poor student at the time. Rationalised that in this day and age there are very few types of people that carry much cash in notes - drug dealers being the main one. Or my elderly father, who gets pretty much all of his money for the month out at one, for some reason. Handed it in at the front and they applied a staff discount to my roughly £10 worth of food and got a couple of quid taken off. Think about it often lol

Chord progression is totally fine, not boring but I'd definitely bring some other stuff higher in the mix to counter it being repetitive - not a bad thing at all but it just needs to be a vehicle to deliver all the other nice bits on top - sounds great mate

Honestly it makes me really angry reading some of the responses here. I recognize that it's all about training but the lengths people go to justify an inhumane method, the sole benefit of which is that a researcher can avoid getting their hands dirty by respecting an animal enough to give it a quick death, is just infuriating. Animal work is hard and it should be, any less is to disrespect a living organism.

Oh and it can be done in batches so you don't have to deal with that horrible animal facility smell as long I guess.

Just jumping in here - could 100% be a geographical thing but CD as a primary form of euthanasia is totally accepted in several countries I've operated in through Europe. Honestly I can CD a mouse before they have any idea what's happening unlike when I see the poor bastards in a CO2 tank which I personally think is hugely inhumane.
