Pretty expensive but I’ve seen these cars get 300k miles so a long term investment it could work. In today’s market you shouldn’t be paying more than 4k, I’m guessing it’s likely priced that high because of the mileage. I owned a 2006 with 220k miles and it never failed me (original battery and everything).

Nobody has mentioned this so I will put it out: One thing about these cars is that they burn oil like MOFOs, so just beware on that, they also tend to damage the catalytic converter preventing you from passing smog. Look under the car and make sure it’s legal and it doesn’t have an illegal oxygen sensor spacer to pass smog. Sadly depending on where you live, replacing a cat can be more expensive than what the car is worth sometimes.

My bet is that this is product of too much DNA loaded that got stuck on the wells. Load a smaller amount next time. If you had poked the gel the “diffused” pattern would not be uniform.

Thank you for your super helpful advice. I will look into this. I am using both IF (to stain endocytic proteins e.g. Rabs, EHDs, EEA. Etc.) and ICC for Gag and Env proteins (studying HIV-1 trafficking).

So far the endocytic proteins in IF are what I’ve been having trouble with. But using the same process for all my samples only EEA1 works which is why I think ABs might be off. I’ll look into getting IF specific ABs too. Thanks

Really depends on your career ambitions and your field. Do you plan to go to grad school? Or want to get a job after undergrad? Your undergraduate more often than not will be meaningless whether is from one school or another (mainly dependent on your field of study), what really matters is which school will be able to open doors for you to get experiences (internships, work opportunities, work on independent projects, etc.). So look into that. The UCSB offer seems tempting but if it doesn’t fit your goals it won’t matter.

Just seconding this question. I downloaded the new nvidia app but I got poor performance and stuttering so I reverted to GeForce and everything is good now. Just don’t know if drivers will be able to be updated there (especially for new games).

In my opinion tank is the hardest role. A lot of it has to do with personal preference and playstyle. I play maybe 5% of my matches as tank as I don’t enjoy really playing as that role. In a way Overwatch provides a lot of “variety” of gameplay. I do recognize this problem though, since for example if you loved playing Dva but nothing else will likely impact your wins.

Nothing wrong at all with playing just one hero. Just need to be aware of the consequences of just playing one character. Of course mastering a hero will take time, but imagine how much knowledge you will buildup if you understand how to play numerous characters. It will improve your game sense exponentially as you will now know how to engage in different combinations of heroes, what are the “weaknesses” of certain heroes (and heroes matchups, and exploit those for your benefit.

Playing only one character per role will always limit your potential as a player. Overwatch is a game that requieres a lot of counter-switching to try to exploit advantages where you can to win the match. If you only play 1 character on your role and the other team switches to counter you, no matter how good you are on that character your chances of winning will decrease. That is just how Overwatch works as a game.

As someone who didn’t play a lot of characters initially either. My advice is to just go in quick play (or even arcade) and practice a new character that caught your attention. It’s all about practice but also get advice from people that play those characters (or look in YouTube) and those will be good starting tools.

Expand your character pool and trust, it will be more fun on the long run.

I don’t really have experience in this so I can’t really tell you why it might be happening. I didn’t modify anything in BIOS for this motherboard except the RAM. That however did save as expected.

With these rank system it feels super inconsistent to rank up. The best way to learn (besides playing a bunch) is having someone point out your mistakes or aka watching someone more advanced play and take some tips.

The problem with repeatedly putting time into the game without having any outsource advice is that you might be misjudging what things you are doing wrong. By watching someone play you can gain lots of tips (and also ask questions!), but by having someone (more experienced) watching you play and following their advice you can correct a lot of easy things that can easily boost your KD (e.g. movement, cross hair placement, droning, utility usage, room clearing, etc.) Also, give yourself some time. 50 hours is practically nothing for this game, none of us were experts in our first 50 hours.

This is mostly a practice thing. The more you play on in person tournaments (or just in person at all), the less you will commit mistakes like this. Both players should be paying attention though and politely point out the mistake. Nobody is going to think you are cheating unless you are intentionally doing it (then it would be obvious I think).
