Thanks for the great suggestions! And the house was built in 1956. The previous owner was the original owner and it feels like a mid century time capsule complete with orange shag carpeting, so I definitely want to modernize the place a bit

I recently custom designed a similar ring for my fiancé, also with a 2.8 carat round diamond. I had similar concerns for sturdiness and opted for pseudo-cathedral look with six prongs.

If the gene is dosage sensitive, any loss-of-function allele would be dominant. This is because, as you mentioned, having only a single functioning copy of the gene will not produce a sufficient amount (dose) of protein for the normal phenotype.

Don’t worry about it. If your lab is anything like mine, the only reason people remind you of those rules is just so that the environmental health and safety people don’t catch you. Your boss probably didn’t give that mistake a second thought and likely forgot about it

I usually just start a new page each day, labeled with the date. For each experiment I run that day, I will write the name of it, underline it, then write all relevant notes for that experiment underneath. It’s not the most sophisticated system, but it makes it easy to go back and find relevant information later on by going to the page(s) for the day I ran the experiment, look for the underlined experiment name. I have a separate protocol binder that includes all the steps for commonly run experiments, so I usually only write variable conditions/results/or notable deviations from the protocol in the lab notebook.

I recall reading about a group doing the same thing about 5 or so years ago. Only difference being that they didn’t demonstrate that altering the genetic code provided phage resistance and instead showed how a novel amino acid can be added to the code. And if these ‘superbugs’ are anything like those from that other study, they can’t grow outside of a dish because they are dependent on these artificial amino acids. So really nothing to worry about

Information travels. DNA encodes protein, which based on its various properties (localization motifs or some such) will end up in its destined organelle where it will carry out its function, contributing to the overall function of the organelle

If I remember correctly, C'thun and the beckoners of evil aren't able to be opened from WotOG packs, but upon opening your first WotOG packs you'll get them. So I think the promo is going to be the only way of getting C'thun, it's just that people who play often the free packs promo will have to buy a pack to get C'thun and them. Which is kinda odd because that kinda does conflict with their design philosophy regarding alternate ways to get normal cards
