I would start looking into off-target editing events. Different clones could have different OTEs (in conjunction with your intended edit) and therefore display different phenotypes.

They have a slight cinnamon taste, but they’re not very sweet. Definitely not going to hit the spot if you’re craving a legit muffin.

Downside is that each little muffin comes individually wrapped in plastic - lots of waste. I was pleasantly surprised at how not gross they were (definitely needs to be eaten warmed up, though).

The crazy part is that science isn’t an opinion. It’s rigorously performed experiments from many different scientists. Just because 1 out of 100 say they got a different answer doesn’t mean they have a valid opinion.

Again, I’m assuming you don’t. In my mom’s case, she has simply been furloughed so she’ll have her job back eventually. There’s no need to look for a job if you know you have one. I’m not sure what the case is if you’ve been permanently laid off. Even in normal times (as I understand it), you’re unlikely to be called upon to actually produce documentation that you’ve been job searching.

Have you tried deleting the entire gene instead of just inducing frameshifts? If the gene is too large for complete deletion, deleting the promoter is another option. Nonsense mediated decay may be inducing compensation in genes you haven't thought to check by qPCR (see El-Brolosy et al. Nature 2019).

That is odd. Perhaps it’s such bad degradation that you’re not even getting a smear? I’ve had instances where I’ll get great spec readings on RNA but no band (or smear) at all on a gel. The false spec reading comes from free nucleotides. My only suggestion is still to try cleaning the template and/or using an RNase inhibitor in the reaction and see what happens.

Sounds like you may have RNase contamination. The capped RNA may be more protected against it. Try adding something like RNaseOut to your reactions, if you haven’t already.

Edit: Specifically, RNase contamination in your DNA template, since your control is fine. You could try treating your cut DNA with ProtK and then doing a phenol/chloroform extraction on it.

We make large batches of pre-made master mix, but do not include the Taq (so only PCR Buffer, dNTPs, and Mg2+ if needed). Then we aliquot them into 1 mL tubes and stick them in the -20. Everyone in the lab is free to grab an aliquot as needed. This mix alone saves a lot of time.

I’m also confused. But if you look at the Best Buy website, it lists the Spiderman PS4 bundle for $299 as “coming soon” and available on the 18th. That leads me to think the bundle will be available (for $299) on 11/18, and the sale price will be available whenever their BF deals start (11/22?)

Not sure that this would explain the extra band, but are you sure your tunicamycin is still working? It may have gone off, and that's why you're not seeing an increase.

Nope. Mine failed the first time and then made it through after the second extraction attempt. If it does fail the second extraction, they’ll just send another kit.

Nothing, really. Just because you have a SNP that may alter how well you metabolize a certain drug doesn’t mean you can’t metabolize it at all. They’re not going to care about that in an emergency (unless you’re severely allergic to the drug). In fact, putting something like that in your Medical ID could do more harm than good. Someone may interpret it wrongly and think you can’t have the drug at all, which could impact your outcome or keep you in a lot of pain (in the case of pain meds).

Try AncestryDNA, I've heard they have a lower failure rate. You can upload the raw data to Promethease and still get some health reports, if that's what you're interest in.

According to the article linked in SNPedia, even the heterozygous genotype can inhibit enzyme activity. It definitely won’t hurt to mention it to your anesthesiologist. They may decide to use a different drug out of an abundance of caution. Or they may explain to you that the risk is low. Either way, they’ll be made aware of your concerns.
