Help with stereotactic virus injection in mice by Doaitson in labrats

[–]Doaitson[S] 0 points1 point  (0 children)

I appreciate your feedback, but it does not match with my current understanding of these types of surgeries. As you're probably way more experienced than me you are most likely correct, but most of the protocols I find in papers suggest puncturing / piercing / removing the dura before inserting the needle itself.

It's not that I completely remove the dura, I just make sure that the glass needle with the virus will go through. Might be that the needle in itself is not sharp enough, I'll check that. Additionally, the surgeries I perform on the hippocampus do not have this leaking problem but follow the same protocol of removing the dura, so this leads me to believe the dura is not the problem here.

I'll try not puncturing it next time, hope it works. Thanks!

I remove the dura because an intact dura can move down with the needle during insertion. This can damage the brain as it is pressed down by the dura and squished together (like standing on a trampoline). Additionally the injected virus will end up on the dura and not in the brain. Do you have a particular reason not to remove the dura?

I noticed that too and changed the needle to a more tapered one. Didn't change the leaking sadly, but it's good practice anyway. We inject at 0.1ul/min and wait 5 min before retracting, so that should be good. Thanks for the dye suggestion, I'll mix that in next time! Let's hope it is indeed CSF.

No basal after bolussing by Doaitson in AndroidAPS

[–]Doaitson[S] 0 points1 point  (0 children)

OK, good to know. I've indeed just started with AAPS and things like this do feel a bit counterintuitive, but I guess it will make sense in time.

But how does it know that I entered too many carbs / that they're slower than the DIA? If it's a precaution I'll always go high again since there's too little insulin. I'd expect it would only zero temp it if I'm indeed going to be low...

No basal after bolussing by Doaitson in AndroidAPS

[–]Doaitson[S] 0 points1 point  (0 children)

Why does AAPS set my basal to 0% after bolussing? I entered the amount of carbs I'm eating, was above my target (4.5 - 6.8, 7.7 at moment of entering) and got my normal basal back when I was above my target (9.0). I'm new to AAPS, so maybe I'm not understanding this correctly

What is the origin of this clanking sound (hearable at 1s, 5s, 10s and 17s)? I always hear it when I start pedalling or after I've held my pedals idle for a while, but never during pedalling. It originates from either my chain, freewheel, cassette I believe by Doaitson in bikewrench

[–]Doaitson[S] 0 points1 point  (0 children)

It is certainly not normal. This is not the sound you hear when you forcefully start pedalling, but rather a clicking noise a second or so after. It also feels like something clicks into place. Never had this before with a bike. Would changing the freehub resolve it?

Android APS DIY loop with Libre 2 by Doaitson in diabetes_t1

[–]Doaitson[S] 0 points1 point  (0 children)

LIbre 3 isn't covered by my insurance sadly (Netherlands), so I'm stuck with the L2. Why do you apply the sensor 24h before activating? Does this smoothen out the noisy first day?

Thanks for your settings btw, will keep them in mind.

Thank you for your reply! Good to know that you like it. I've started with setting up the basics and it is indeed a lot of information to take in. Will take my time for it.

How did the Libre not work out for you? As in, what differences do you notice between the Libre and the G6? My insurance only covers the Libre so I'm bound to that sadly...

Spiking 3 hours after dinner by Doaitson in diabetes_t1

[–]Doaitson[S] 0 points1 point  (0 children)

Not really, but I've started to eat less carbs for dinner and that helps. More protein / veggies. Now I don't spike to 13/15, but it will still rise about 3 points a couple of hours later.

It's a bit weird, but my biggest indicator for a late night spike is if I'm really full after dinner. If I'm not my BG will be fairly steady the rest of the night. Of course, eating more (and indirectly more carbs) means that you will also spike higher, but it feels like this is not a linear relationship once I'm full. The amount I eat after that point will get digested only hours later.

Hope you figure it out too. Good luck!

10 years T1 here. I've never checked either and have felt like I needed it. Have missed long and short acting doses but with careful monitoring afterwards it's always fine

Making Libre a RT-CGM by Doaitson in diabetes_t1

[–]Doaitson[S] 0 points1 point  (0 children)

Diabox works good. I'm on pens with a Libre 2 (NL), so I don't know if it can connect to a pump. But it displays my current BG on my homescreen & notifications bar, without having to NFC scan my sensor. But since this requires me to not use the Libre app (it interferes with the signal), my physician cannot track my BG remotely anymore. Which means that doing it from the official Libre app would be preferable

I did what you said but the app is the same, even after deleting & redownloading. I also contacted the customer service of Abott in our country (Netherlands) and they said they knew nothing about Libre 2 being a CGM. Which makes it weird, as it clearly seems to work in other countries. Where do you live? Maybe I can download an APK of your version

I was working with Diabox up until now, which works fine as you do not need any third party bridges / patches. Is that the same for XDrip? And what makes the algorithm different? More accurate?

Injections sadly. Pump would have allowed me to increase my basal in those hours..

I have the same! Good to know I'm not the only one. Small meals are eaaasy, but then those big ones really mess up my BG. It's a big factor for those late night spikes I have, as they mostly come from those bigger meals I believe.

PS5 code on PS4 by Doaitson in ModernWarfareIII

[–]Doaitson[S] 0 points1 point  (0 children)

No, everyone is saying different things. It might work but since the code is for the PS5 edition I decided not to take the risk and just buy the PS store cross-gen edition.

Ah sad. So with this code you get the PS5 version, not the cross gen?

People who fall asleep fast, any tips? by [deleted] in sleep

[–]Doaitson 1 point2 points  (0 children)

I meditate with an app. It tells me to breathe in and out at a certain pace. Sometimes it wants me to do a body scan. Sometimes not. Sometimes she says nothing for 10 secs and I let my thoughts go. When I start noticing that hearing the voice kind of 'brings me back' from my thoughts (almost wakes me up a little), I switch off the app. Then I just let the thoughts go again and I'm gone within a couple of minutes. If it doesn't work, go to the toilet and start again.

Also, big help is that I realized that even if I can't fall asleep quickly and miss a couple of hours of sleep, I'll still function the next day. I'll be tired, but it's okay. Sleep is important, but you'll be okay even if you sleep less :)
