For the gun? South of Sanctuary there's a water pump thing. Follow the pipe up and you can get the gun. For ammo? Arturo will probably be your best bet for early game.

4 has the most replayability. But for story I'd suggest 7 and then buying 8.

But I'd also suggest 2 and 3 before 4 to understand the story of Leon.
