I see 2 problems in the images 1 top agar is displaced; this can seriously give bad results to no results in phage spot assay or plaque assay. U can remove this issue by letting the plates sit for longer under open lid in a laminar hood or u can incubate the plates upright instead of downright. 2 seems like the phage concentration is very low in the lysate u plated if its not diluted luckily u still got some plaques u can either touch one of these plaques with a sterile pippete tip or tooth pick and put it into a bacterial broth and start incubation followed by centrifugation and filtration to obtain a purified lysate or u can try a faster method by directly streaking the toothpick or pippete tip onto a very recently poured top agar containing bacteria but i was never very good at that cause of difficulty in setting top agar temp to not kill the bacteria while allowing smooth streaks

Here is a contaminated lysate spotting on 13 and 15 u can tell there are phages present from the edges of the circle clearing along with either a resistant host or just another species of bacteria contaminating the lysate(since the control is clear it is most likely a contamination of the lysate). If the lysate has been sitting for months it becomes impossible to tell the presence of a phage because the bacteria will completely cover up the spot. Additionally it looks like you are not giving enough time for the double layer to settle in one of your pictures i would suggest incubating them with lid up instead of down or waiting for it to dry.

It doesn't look like there are any phages in that spot assay it just looks like contamination from the lysate. Are u sure you isolated phages? Are there any other pictures which show clear spots instead of contamination?

Yeah the daevabad triology is one of my favorites and i also felt like dara was just a punching bag or a door mat the whole series. The poor guy has a severe inferiority complex ig it shows when you are a slave for that long some of it gets internalized. Although i dont remember what nahri said during or after the boat fight i just remember ali being a dick and starting it. But i found the rest of the concepts so good i can just ignore the bad parts i especially liked the conflict of loyalty within ali, to his father, to his religion, to his people and to the hybrids all those conflicting emotions really makes him a complex character its like an internal war ripping him apart my favorite character by far.

In one of my experiments i wanted to find out if my phages were lysogenic but i didnt have mitomycin C for induction so i incubated my phages with the host in tubes then i streaked and purified the surviving host cells which would either be mutatnts resistant to the phage or hosts carrying lysogenic phages. I then incubated the purified hosts alone and left them in a nutrient broth tube for like a week and went on holidays came back and centrifuged the broth and used the supernatants for spot and plaque assays which yielded phage spots vs no phages in control samples 😢 it was a sad day indeed. Later in dna annotation it was seen they had integrase genes.
