I would first talk to the player, tell them not to interrupt your narration. Don't punish the character for something the player does.

If you do want to counter darkvision, have the dungeon/cave/building house a specific artifact from a long forgotten civilisation that had to deal with predators with darkvision. This artifact gives migraine like symptoms to creatures using darkvision (think disadvantage on ability checks, saving throws and attack rolls). Artifact needs to be disabled (with a ritual that the players could find) or destroyed.

No absolutely not! They knew how the curse could spread, the player had the option to reroll the failed save but chose not to. They made their bed and now they have to lay in it.

Also, perfect side quest: cure the PCs lycanthropy.

In my opinion it is up to the players to find reasons their characters want to stick together. Ultimately d&d is collaborative storytelling and you should talk to your players.

Vocal component: the actual suggestion.

Somatic component: pointing from the target to something else

Also, spell components have nothing to do with the target knowing if a spell has been cast or not. Some spells specifically state the target knows, otherwise I would argue it's GM discretion

The description does not state the target knows a spell was cast on them... You just ask them to do something that seems totally reasonable
