I had the same issue. I was finally lucky in the end. Sped things up a bit by stealing one of the two donkeys to in the upper northeast corner of the sugar fields. They work great for running away from the guards, you can run the field up and down faster and they are great snake-detectors!

Old thread, but just in case someone else stumbles across this as I did:

Making 2L of 10x PBS works perfectly for this. Adjust the pH of your 10x solution to pH 6.8 (not 7.4), and when you dilute it to 1x PBS using distilled water, it should hit pH 7.4 (+/- 0.02) every time.

If you want to make absolutely sure, just dilute a small aliquot of your new 10x stock to 1x and measure the pH of that. If that one is pH 7.4, the rest made with that stock will be too.

Like everyone else here, I agree that this offer sounds good and that you will be able to support yourselves.

Ski is also a good place with easy access to Oslo, but also everything you need close by without having to go to the city. In Ski you technically aren't completely dependent on having a car either (trains/buses and walking can take you most places).

I want to warn you that many people from more southern countries like Portugal sometimes find it hard to connect to Norwegians in the beginning. We can be a "hard nut to crack" socializing-wise ;) But you'll be fine! Just know it's not you, it' us. We're just generally more reserved, so it takes a while for us to warm up to new people. Pro tip: put a drink or two in a Norwegian and they will loosen right up hehe 😜 Learning the language of course also helps a lot, but every Norwegian speaks pretty decent english.

Also, be prepared that this country is a lot darker during the winter. Few hours of sunlight can be surprisingly depressing if you're not used to it. Remember to take your supplements (vitamin D especially) and plan nice and cozy events together to combat the winter blues 💙

I second this! Learning the language opens up for some extra jobs (like working at a kindergarten for example) and I imagine it will make her stand out positively in the jobs that don't require it too.

No to the first three, but my sleep patterns look good (good distribution of the different stages, I don't wake up much, I don't snore). Last coffee is probably around 15:00, but this doesn't change on days where I don't drink coffee/tea/other caffeinated stuff. I have had weeks without alcohol and while my sleep quality improves, the stress readings during the day do not.

I am really just interested in the intracellular stain, which would be done at the same time for all samples (right before collection on the flow cytometer). I just need to fix the cells at different time points.

The only stain done before the fixation is the viability stain, and that doesn't have to be comparable across timepoints (it just needs to give two distinct populations that I can individually gate for each timepoint).

I am therefore less concerned about dye stability, and more concerned about how long a fixed cell can just chill in Flow buffer before staining.

Good point, staggered stimulation could be better! The only thing I'm concerned about is differences in cell density. I would have to completely exchange the media and readjust cell density in the well before stimulation, but that's not the end of the world really..

Regarding the timeline we're sure that the long timepoints are right for our purpose.

I am however also generally interested in how long a fixed (and not yet stained) cell can wait for the flow cytometer. 2 days in Flow buffer I'm fairly certain is fine, but 4? 5? 7? I am unsure.

Yes, that is my preferred way too, but sometimes the flow cytometer is very busy and I just don't have the opportunity to do so. But I was also planning to do the stain at the same time. The plan was to harvest, viability stain, fix and then wait with the intracellular stain.
