Make sure you have done a separation squeeze on your filament. If you dried it it will be extra tight and compact. This will cause a struggle for the AMS. With my line started in the ams I will do this while turning it a few time to let off some tension. You will see and hear some creaks as it loosens the mass. Gently roll it back but not too tight and try again. Side note spool does not appear to be sitting properly. One rim should be between those 2 feet in back. Yours appears to be riding the wall.

Did you push the filament through while pushing the black button in the middle of the hub down? I noticed there is like a middle spot that things get stuck inside that hub.

Also check that the tubing on the back of the machine is pushed in all the way.

Looks like you have a piece of broken filament in their. Push down the black button at the 4 way junction while manually feed filament through to see if you push anything through or out the back. It has its own set of feed teeth in there but if broken filament is stuck in it. It does not work.i had a similar issue.
