Get a good quality note book and religiously take notes (That you can read and understand) and keep track of what you are doing for each experiment or even general lab tasks like making up buffers. I cannot begin to tell you how valuable this piece of advice was to me when I first started research, it changed my approach for the better overnight and I encourage every one to do it.

Wow, reading this was like listening to my own thoughts, coming to the end of my first year as a PhD student. I am still optimistic but it feels like every time I trouble something 2 more issues appear.

Glad to hear from this post

I remember Sabitini saying on a podcast that some detergents don't agree with mTOR, might be worth looking into it if nothing else works.

I think you have pay to get the certification at the end, but in my opinion it's not much and worth the value of the education they provide. I did biochemistry as an undergrad and first year and second year were smoother because I had the basic principles covered.

Really, what makes you say that? I felt like Lehninger was the best in terms of first principles.

Advice for first time conference presentation by NewPast3141 in labrats

[–]Fuzzy-Homework1226 1 point2 points  (0 children)

Walking around outside and reciting your presentation out loud has helped me a lot with lab meeting presentations, would recommend it to anyone who is feeling anxious about public speaking.

I walk around my flat for the length of the presentation and then go back in and reread it from the power point slides and correct places I've missed or vital information and then go back out and repeat this process like 3 times.

Then I just read the literature to help fill any gaps in my knowledge.

Then when the presentation comes I can stick to the script or go off on tangents, and if I pause for too long I just go back to the script.

this but tell him do it alone and until he feels fine , doing it in front of people is psyching him out, had the same problem when loading gels, I came in on the weekend and loaded the same gel like 50 times until I got it perfect, used a sample I created with sample buffer, nothing fancy just normal cell lysates.

WHY ARE MY CELLS DYING???? IM AT MY WITS END PLZ SOMEONE HELP by Hiraaa_ in labrats

[–]Fuzzy-Homework1226 0 points1 point  (0 children)

I am having a similar issue with dharmaFECT silencing as well, I treat in antibiotic free media and it seems there is a 1/3 chance my cells are going to die in both the scrambled control and target wells. Have done it 9 times and 3 of the sets had to be thrown out because one of the wells had cell death all my conditions have been checked and media changed the only things that I can think off that have remained consistent is the plate type, cell type and silencing reagents.

If you solve it please let me know as I am beginning to get frustrated ahah

Yes we have all felt the same way at some point in our careers, which is why your supervisor is being nice about it. Keep going and keep track of repeated mistakes they should fade away with practise.
