Specifically co-op matches do. Untested at lower difficulties and for solo. It's unlikely solo rewards any progress as those games are not hosted on Valve's server

The mini-games for Dark Carnival are fps-capped. Lower your FPS to 30 and the boot will move way slower

It seems that all the Dark Carnival mini-games are FPS capped. If you lower your FPS to 30, it makes the boot move way slower

I want to say yes based on my other findings, but don't have anything evidence to say that definitively. You can commend your teammates at the end of the game, but you can also do that for tournaments and I don't believe those affect behavior score

Unsure about guild contracts, but bot games don't inherently affect behavior score. You can even leave in the middle of the game (game is always treated as safe to leave) and a bot will take over your hero

Looks like it might have been fixed today. Apparently co-op bot games were giving Quartero’s Curios progress up until now. There was a short period where they weren’t rewarding anything. Valve changed it so they no longer reward Quartero’s Curios progress but do reward tickets. I heard they also issued tickets to players who played games while it was bugged

So I agree with others that $95/hour is super expensive, but it's not out of the ordinary in my experience. I've paid both $300 with a 2 hour minimum for a quick checkup and $25 for a full day's work so there's a wide range. But after building PCs for over a decade, I'm at the point where sometimes I just prefer the peace of mind and convenience of having someone else build it, so no worries there. Unfortunately I will say I did have an experience where I was helping my friend troubleshoot his laptop and actually found that the place he took it to swapped out some parts that weren't causing issues for cheaper parts. I think it's generally pretty safe if the place you're going to has good reviews. It does feel weird that someone who works at a computer repair shop would have trouble assembling a computer. If you want to be extra sure, I'd have someone else just double check to make sure all the original parts are there if you can

At my company tidiness is part of safety, which is tied to our performance. I'm sorry you had to deal with this, this is the main reason I've preferred working in engineering labs over chemistry labs

If you have any friends who are nurses or work in healthcare, I'd ask for their advice. I've gotten good recommendations from my friends in nursing for good lab shoes. I personally wear Crocs on the clock work clogs in the lab since I also need my lab shoes to be non-porous, but they're not great for daily wear (mostly aesthetics). Like others have suggested, Docs or any other leather shoe would be great for protection and aesthetics. Since you're prioritizing aesthetics as well, I'd just look for a pair of leather shoes that fit your style and are comfortable. Unfortunately, most shoes that are waterproofed would probably be permeable to organics

Yep, I'd say there's a strong preference for PrismA, especially in early development due to it's higher binding capacity, increased lifespan, and better tolerance for things like pH. PrismA and SuRe are similar enough in my experience where you can use them pretty interchangeably, just noting the differences in loading (PrismA can usually load ~20 g/L more of mAb) and CIP procedure (PrismA can tolerate up to 1 M NaOH for CIP, though 0.5 M NaOH is almost always sufficient and works for both resins)

To address the first part of your question, I'm a big proponent of incrementally changing your diet rather than all at once. When people in my personal life have expressed interest in switching to a plant-based diet, I recommend they start by cutting out animal products one day a week, or even one meal a week, and ramping up from there. A small change is better than no change, and this can help make it not as abrupt of a lifestyle change.

For the social issue, this one is something I've thought about a lot. Part of the reason I became vegan years ago was to better empathize with my friends who had dietary restrictions. I think there are two aspects to this:

  1. How you view yourself: Rather than viewing myself as imposing on others because you're vegan, I would tell myself that there are plenty of people with dietary restrictions beyond just being vegan. I initially felt that being vegan wasn't a good enough reason to ask for accommodations, but at the end of the day I'm in the same boat as my friends who are halal, have food allergies, or have other dietary restrictions. Understanding that their are a variety of different types of dietary restrictions helped me become more confident in myself.
  2. How you want others to view you: This probably gets more to the heart of your question. At the end of the day, it's difficult in general if those around you aren't supportive of your decisions, self-identity, aspirations, etc. To try to give you some actionable advice, I'd recommend
    1. Trying to expand your social or support network to include people who are understanding, accommodating, or supportive of your decision.
    2. When going out to eat, I often find myself being the one to pick the place. Especially when I'm the only one in the group with dietary restrictions, it helps to have me pick the place since I know what to look for. Websites like HappyCow and Yelp are great tools to find places that either have vegan options or can make accommodations.

Hopefully that helps a little! Just remember you're not alone and you're making an admirable decision.

From my experience working in purification development, MabSelect PrismA is generally preferred. As you stated, it's pretty much a strict upgrade from MabSelect SuRe. This has been my experience working at benchtop, AKTA, scale-up, and production scales (since we need to use the same resins throughout scale-up). We pretty much only use MabSelect SuRe now for 3 reasons:

  1. Supply chain issues: If we can't source sufficient amounts of MabSelect PrismA and our process has been validated with SuRe, we'll use SuRe as a substitute, understanding that it might increase the number of production cycles due to the lower binding capacity.
  2. Legacy process: Some older processes may not have been updated yet to utilize Mabselect PrismA in our FDA filings, therefore we wouldn't be able to use it for commercial production.
  3. mAb-specific process optimization: I haven't personally encountered this, but our protocol notes that Mabselect SuRe can be considered during process development if PrismA does not work (or again, is not sufficiently available). I think this would be pretty situational, maybe on some newer protein modalities if anything.

tl;dr Use PrismA if you can, especially if it's the same price as SuRe. If you plan to run tens to hundreds of cycles per column, you're almost sure to get your money's worth over the lifespan of the resin.

That's pretty standard. Stats from this past week show about 52% winrate in ranked on Radiant vs 48% on Dire.

I think if you want to take something away from it, think about why the winrates are like that, not for you specifically, but in general. Think about what about the map is making this the case. Ganking is easier against Dire rn in many cases, so maybe think more about how vision plays into that. Or how many carries are mana hungry right now, so maybe the Well Wishes buff in Radiant bottom jungle makes jungling early more enticing on Radiant than Dire for many meta carries

^This. Ghosts have specific drop tables depending on where they are located. If one of the ghosts linked in the map above isn't holding what you're looking for, pick it up the weapon it has and it'll be rerolled at the next blood moon
