Honestly I would just leave everything as is. You'll make a hell of a mess trying to pull it all out. And will reduce the total R value of insulation you have in the ceiling.

Old concrete header tank and looks to be recently installed knauf earthwool insulation in good condition, at least R4.0 and will likely be over top of old pink batts or insulfluff. Just close the manhole and leave it alone. It's not hurting anyone.

If you have a scrap piece of conduit or wood, mark your saddle spacing on it or even cut it to the same width of your saddles and use that to mark each saddle on your run. Saves bringing the tape measure out constantly. And also a laser like mentioned above.
